Francisco Q. Goncalves et al.

Summary: The increased release of ATP and the subsequent formation of extracellular adenosine, which activates A(2A)R, play a key role in the abnormal synaptic plasticity associated with the onset of motor symptoms in Parkinson's disease.

Extracellular ATP: A powerful inflammatory mediator in the central nervous system

Francesco Di Virgilio et al.

Summary: Nucleotides, especially extracellular ATP (eATP), play a crucial role in inter-species extracellular signaling. Different organisms have evolved sensors with varying affinity and selectivity for eATP, resulting in a highly flexible signaling system. In mammalian evolution, the control of intracellular ATP and eATP homeostasis is closely linked to that of Ca2+. Specific plasma membrane receptors, known as P2 receptors (P2Rs), sense eATP with various degrees of affinity and selectivity, adding to the plasticity of the system. The central nervous system (CNS) is a privileged site for purinergic signaling, with all brain cell types expressing P2Rs. Evidence suggests that eATP not only participates in synaptic transmission but also plays a crucial homeostatic role in brain cell responses. Modulating eATP concentration in the CNS may hold promise for the treatment of neuroinflammation.

Anderson Camargo et al.

Summary: This study reveals the antidepressant-like effect of Guanosine and the modulation of A(1)R and A(2A)R in its antidepressant action. Guanosine interacts with A(1)R and A(2A)R at the adenosine binding site.

The ATP Level in the Medial Prefrontal Cortex Regulates Depressive-like Behavior via the Medial Prefrontal Cortex-Lateral Habenula Pathway

Song Lin et al.

Summary: This study systematically investigated the cellular and neural circuit mechanisms underlying ATP deficiency-induced depressive-like behavior. The results showed that the deficiency of extracellular ATP led to reduced inhibition and elevated excitability in lateral habenula-projecting medial prefrontal cortex (mPFC) neurons. The P2X2 receptor in GABAergic interneurons mediated ATP modulation of depressive-like behavior.

Adenosine A2A receptors control synaptic remodeling in the adult brain

Xinli Xu et al.

Summary: This study reveals that adenosine A(2A) receptors (A(2A)R) contribute to circuit remodeling in the mature brain, specifically in the hippocampal mossy fiber (MF) sprouting found in diseases like temporal lobe epilepsy. A(2A)R activation in dentate granule cells induces the formation of abnormal secondary axons, which is activated by extracellular ATP-derived adenosine. These findings highlight the role of the CD73-A(2A)R axis in regulating circuit remodeling in the mature brain.

Convergence of adenosine and GABA signaling for synapse stabilization during development

Ferran Gomez-Castro et al.

Summary: The research demonstrates that activation of adenosine A(2A) receptors is essential for stabilizing GABAergic synapses during neural circuit development. The synergistic action of GABAAR- and A(2A)R-dependent signaling pathways leads to the production of adenosine 3',5'-monophosphate. The stabilization of pre- and postsynaptic GABAergic elements involves the interaction between gephyrin and the synaptogenic membrane protein Slitrk3.

Crosstalk Between ATP-P2X7 and Adenosine A2A Receptors Controlling Neuroinflammation in Rats Subject to Repeated Restraint Stress

Liliana Dias et al.

Summary: This study demonstrated the functional interaction between P2X7R and A(2A)R in regulating microglia reactivity in response to repeated stress, indicating a potential cooperation between these two purinergic systems in controlling brain function. The results show that antagonists of P2X7R and A(2A)R can attenuate the behavioral modifications caused by repeated stress, and their effects on calcium levels in microglial cells suggest a complex interplay between these two receptors.

Increased ATP release and CD73-mediated adenosine A2A receptor activation mediate convulsion-associated neuronal damage and hippocampal dysfunction

Summary: The study demonstrates that ATP release, CD73, and A(2A)R density increase upon convulsive activity, playing a key role in neurodegeneration. Genetic deletion of CD73 attenuates neuronal degeneration but has no impact on astrocytic modifications. Furthermore, knocking out CD73 eliminated the deterioration of long-term potentiation and memory performance caused by convulsions.
