Catechol-Based Polymers with High Efficacy in Cytosolic Protein Delivery

Summary: This study demonstrates that polyphenols can serve as highly efficient carriers for the delivery of macromolecular biopharmaceuticals. Catechol-modified dendrimers show the highest efficacy for protein delivery, while pyrogallol-grafted ones have the lowest efficacy due to increased hydrophilicity. These catechol-based polymers efficiently deliver bioactive proteins into the cytosol of live cells and have the potential to inhibit tumor growth in vivo.

Fluorination Promotes the Cytosolic Delivery of Genes, Proteins, and Peptides

Summary: This study proposes the use of fluorous ligands to promote cytosolic delivery of biomolecules. Fluorination significantly improves gene delivery efficacy and serum tolerance of cationic polymers. Additionally, fluorinated polymers are efficient for intracellular protein delivery and cytosolic peptide delivery, outperforming traditional techniques.

How Do Biomolecules Cross the Cell Membrane?

Summary: This paper introduces a previously unrecognized membrane translocation mechanism called the vesicle budding-and-collapse (VBC) mechanism. Through VBC, biomolecules can autonomously translocate across cell membranes. The study found that CPPs and bacterial protein toxins can escape endosomes through the VBC mechanism, and proposed that VBC may be the mechanism that drives the bacterial TAT and eukaryotic UPS systems.
