NIR-driven intracellular photocatalytic oxygen-supply on metallic molybdenum carbide@N-carbon for hypoxic tumor therapy

Summary: The metallic Mo2C@N-carbon@PEG nanoparticles were developed for generating near infrared (NIR)-photocatalytic O-2 and enhancing photodynamic therapy (PDT), showing potential for anti-cancer therapy integration. The nanocomposites exhibited high NIR absorption and photothermal conversion efficiency, with enhanced performance compared to pure Mo2C. The novel biodegradation and metabolism behaviors of the nanocomposites were also investigated, demonstrating elimination from the body within 14 days.

Summary: Photodynamic therapy (PDT) has limitations for deeply embedded tumors due to poor light penetration. Chemiluminescence resonance energy transfer (CRET) provides a promising alternative to PDT without external light. However, using endogenous hydrogen peroxide (H2O2) and oxygen (O-2) to trigger CRET-mediated PDT restricts treatment efficacy. In this study, a nanohybrid nanoparticle (NP) was designed to achieve synergistic CRET-mediated PDT and calcium (Ca2+)-overload-mediated therapy against hepatocellular carcinoma.

Summary: A stable and efficient in-situ oxygen self-supply system for hypoxic tumors is proposed using organometallic drug complexes with a supramolecular interaction strategy. The ruthenium (II)-coordinated supramolecular metallodrug complex (RuSMDC) is designed to produce oxygen by catalyzing the decomposition of hydrogen peroxide, leading to improved therapeutic effects of chemotherapy and photodynamic therapy oncotherapy in anaerobic tumor environments. Further studies confirm that Ru-containing supramolecular metallodrug micelles effectively alleviate the tumor hypoxic environment and reduce systemic toxicity.
