Mechanistic Models for USP2 Dissolution Apparatus, Including Fluid Hydrodynamics and Sedimentation

Physico-chemical characterization of aspirated and simulated human gastric fluids to study their influence on the intrinsic dissolution rate of cinnarizine

Summary: This study investigated the critical parameters affecting drug dissolution in the human stomach by determining the intrinsic dissolution rate of cinnarizine in aspirated and simulated human gastric fluids. The results showed that the pH dependent drug solubility and viscosity differences of the simulated gastric fluids significantly influenced the dissolution rate of cinnarizine. Using a medium with viscosity similar to water to evaluate gastric drug dissolution may overestimate the dissolution rate. The developed simulated human gastric fluid showed promise in studying gastric drug dissolution in vitro.

An Enabling Formulation of a Weakly Basic Compound Guided by Physiologically Based Biopharmaceutics Modeling (PBBM)

Summary: Weakly basic compounds may precipitate out of solution when passing from the stomach to the intestine, reducing drug absorption. Gastric pH and precipitation time were found to be influential factors for absorption, while particle size and gastric residence time had no impact. By selecting appropriate formulation and adding specific excipients, the risks associated with intestinal absorption can be minimized.

Exploring precipitation inhibitors to improve in vivo absorption of cinnarizine from supersaturated lipid-based drug delivery systems

Summary: The study explores the use of precipitation inhibitors in supersaturated lipid-based drug delivery systems to enhance the bioavailability of cinnarizine. Results showed that inclusion of precipitation inhibitors led to increased cinnarizine bioavailability, particularly in certain formulations. By mitigating the risk of in vivo precipitation of cinnarizine, precipitation inhibitors allow for comparable bioavailability to non-supersaturated lipid systems.
