Validation of an IFN-gamma ELISpot assay to measure cellular immune responses against viral antigens in non-human primates

Fan Yang et al.

Summary: The study successfully validated the IFN-gamma-based ELISpot assay for measuring T cell responses against viral antigens in cynomolgus monkeys, but monitoring peripheral T cell responses in non-clinical AAV5 gene therapy studies was found to have limited value for interpreting safety or efficacy.

Gene Therapy Advances: A Meta-Analysis of AAV Usage in Clinical Settings

Hau Kiu Edna Au et al.

Summary: Adeno-associated viruses (AAVs) are the safest and most effective gene delivery vehicles in gene therapy, but their translatability into clinical settings is partly limited. This study analyzed over two decades of clinical trial data and found variations in the speed of novel AAV development in different therapeutic areas. More clinical data on dosing regimes, immune-modulation strategies, and gene editing approaches are needed to accelerate the translation of AAV-mediated gene therapy.

Comprehensive evaluation of the effects of long-term cryopreservation on peripheral blood mononuclear cells using flow cytometry

Bo Li et al.

Summary: This study used flow cytometry to investigate the effect of long-term cryopreservation on the quality of peripheral blood mononuclear cells (PBMCs), and found that long-term cryopreservation did not affect the recovery and viability of PBMCs, but led to a decrease in the number of innate immune cells and dynamic changes in the proportions of T cell subtypes. This study provides important evidence for the effective utilization of biobank resources.

Summary: Gene transfer using AAV vectors has made significant progress, but is still hampered by immune responses and high doses may lead to severe adverse events. More efficient AAV vectors and alternative dosing strategies should be explored.

Immunogenicity of Novel AAV Capsids for Retinal Gene Therapy

Miranda Gehrke et al.

Summary: This study compared the immune responses of new AAV vectors with the parental AAV2 vector. The results showed that the new vectors were less sensitive to neutralizing antibodies and had similar cellular immune responses and cytokine production compared to the parental vector. Due to their enhanced retinal tropism, the new vectors are likely to have lower immunogenicity for gene therapy.

Immune responses to retinal gene therapy using adeno-associated viral vectors-Implications for treatment success and safety

Kirsten Bucher et al.

Summary: Recombinant adeno-associated virus (AAV) is a leading vector for gene therapy in the retina, efficiently transducing retinal cells but also causing intraocular inflammation and immune responses. The eye, being an immune-privileged site, may mitigate these responses, but further research is needed to understand influencing factors.

Summary: The evaluation of immune responses before and after AAV-mediated gene therapy plays a key role in determining therapeutic safety and efficacy. This study found that administration of BMN 270 mainly led to antibodies against the AAV5 capsid, with no FVIII inhibitor responses observed within 3 years of treatment.

Understanding Early-Life Adaptive Immunity to Guide Interventions for Pediatric Health

Eleanor C. Semmes et al.

Summary: Understanding the distinct nature of the infant adaptive immune system is crucial in guiding immune-based interventions, such as vaccines, in early life. The differences in T cell, B cell, and humoral immunity between infants and adults hinder the development of long-lasting immune responses in infancy. Applying this knowledge to optimize vaccine strategies and integrating maternal and infant immunization approaches can enhance neonatal immune protection.

Summary: AAV-based gene therapies show promise for treating hereditary diseases, but may trigger cellular immune responses.
