Electrospun PLGA and PLGA/gelatin scaffolds for tubularized urethral replacement: Studies in vitro and in vivo

Summary: The study investigated the biocompatibility of PLGA and PLGA/gelatin scaffolds for tubular urethral replacement in a canine model, finding that while both scaffolds supported urothelial cell growth, they were not suitable for urethral replacement due to varying degrees of urethral strictures postoperatively. The PLGA/gelatin scaffold had increased tensile deformation and suture retention strength compared to PLGA. Postoperative recovery was good, but there were issues with urethral stricture and lack of continuity in multilayer urothelial cell formation in the reconstructed urethras.

Urinary bladder augmentation with acellular biologic scaffold-A preclinical study in a large animal model

Summary: The study evaluated the utility of bladder acellular matrix (BAM) for reconstructing large urinary bladder wall defects in a long-term porcine model. Six out of ten animals survived the 6-month follow-up, with four dying due to scaffold failure or other complications. Reconstructed bladders showed normal function with lower expression of urothelial, smooth muscle, blood vessel, and nerve markers compared to the native bladder.
