Gene by environment interaction mouse model reveals a functional role for 5-hydroxymethylcytosine in neurodevelopmental disorders

Summary: Mouse knockouts of Cntnap2 result in altered neurodevelopmental behavior and deficits in striatal GABAergic signaling. Additionally, it leads to a disruption of DNA methylation modification in the orthologs of genes implicated in human neurodevelopmental disorders. Prenatal stress can induce repetitive behaviors and altered sociability in Cntnap2(+/-) mice, similar to the homozygote phenotype. The changes in phenotype are correlated with disruptions in hippocampal and striatal 5hmC levels and altered transcript levels of related genes.
