Mapping of Nonhomologous End Joining-Mediated Integration Facilitates Genome-Scale Trackable Mutagenesis in Yarrowia lipolytica

Xiaoqin Liu et al.

Summary: The study revealed the distribution of NHEJ-mediated integration on chromosomes for the first time and demonstrated its potential for constructing a genome-scale trackable mutagenesis library. The research showed that the integration inserts DNA randomly across chromosomes and exhibits a preference for transcriptional regulatory regions. This approach proved to be effective in rapidly identifying new targets for improving beta-carotene biosynthesis and acetic acid tolerance.

Biotechnological advances for improving natural pigment production: a state-of-the-art review

Xiaomei Lyu et al.

Summary: Due to the increase of people's health awareness and the discovery of novel pharmacological effects, natural pigments are experiencing a rapidly growing global market. However, traditional production methods are limited by the source and low content of target pigment compounds. To achieve industrial-scale production, many efforts have been made to increase pigment production through the development of in vitro plant cell/tissue culture systems and optimization of microbial cultivation methods. Synthetic biology has also provided new approaches for the biosynthesis of pigments from different sources. This review comprehensively summarizes the innovative methods and strategies for optimizing and engineering both native and heterologous producers of natural pigments, presents current progress in the production of several representative high-value natural pigments, and discusses the remaining challenges and future perspectives.

Targeting pathway expression to subcellular organelles improves astaxanthin synthesis in Yarrowia lipolytica

Yongshuo Ma et al.

Summary: This study explores the strategy of expressing the astaxanthin biosynthesis pathway in sub-organelles of the oleaginous yeast Yarrowia lipolytica to achieve efficient synthesis of astaxanthin, leading to increased yield and reduced accumulation of intermediates. Targeting the astaxanthin pathway to subcellular organelles accelerates conversion and significantly improves synthesis, ultimately enhancing compartmentalized isoprenoid biosynthesis in Y. lipolytica.

Astaxanthin for the Food Industry

Barbara Stachowiak et al.

Summary: Xanthophyll astaxanthin is an expensive and important industrial pigment commonly used in aquaculture, responsible for the pink and red color of salmonid meat and shrimp. It has strong antioxidative properties and health benefits, thus being utilized in nutraceuticals and cosmetics. However, its application in food technology is limited due to concerns about the effects of synthetic astaxanthin on human health and the high cost of natural astaxanthin.

α-Farnesene production from lipid by engineered Yarrowia lipolytica

Yinghang Liu et al.

Summary: This study explored the ability of Yarrowia lipolytica to synthesize alpha-farnesene, an important sesquiterpene, using lipid feedstock. By increasing the copy number of key genes and introducing a heterologous gene, alpha-farnesene production was significantly improved. The optimization of metabolic engineering and fermentation conditions led to a successful synthesis of alpha-farnesene with high titer and yield from waste lipid feedstock.
