Expanding the Scope of Polyoxometalates as Artificial Proteases towards Hydrolysis of Insoluble Proteins

Nada D. Savic et al.

Summary: The study reports the hydrolysis of fully insoluble protein using polyoxometalate complexes as artificial proteases in surfactant solutions for the first time. The results indicate that the hydrolytic selectivity and activity of the POM catalysts strongly depend on the concentration of surfactant.

The Dawn of Metal-Oxo Clusters as Artificial Proteases: From Discovery to the Present and Beyond

Francisco de Azambuja et al.

Summary: The selective cleavage of peptide bonds in proteins is crucial in biological and medical sciences, with current applications relying on costly proteases like trypsin. Transition-metal complexes, providing larger fragments and complementary information, have emerged as selective artificial proteases. Their selectivity profile is determined by various structural factors, offering a promising alternative to traditional proteases.

Broadening the Scope of Polyoxometalates as Artificial Proteases in Surfactant Solutions: Hydrolysis of Ovalbumin by Zr(IV)-Substituted Keggin Complex

Nada D. Savic et al.

Summary: The development of catalysts for selective protein hydrolysis is crucial for various biotechnology and proteomics applications. This study investigated the proteolytic potential of catalyst systems based on Zr(IV)-substituted Keggin polyoxometalate and three different surfactants. The presence of surfactants influenced the performance of the catalysts, but did not affect their selectivity. Interactions between OVA, surfactants, and Zr-K 1:2 were explored using techniques such as tryptophan fluorescence and Circular Dichroism.
