Ileal Bile Acid Transporter Inhibition Reduces Post-Transplant Diarrhea and Growth Failure in FIC1 Disease-A Case Report

Johanna Ohlendorf et al.

Summary: Familial intrahepatic cholestasis 1 (FIC1) disease is a genetic disorder that often requires liver transplantation. Post-transplant patients may experience severe refractory diarrhea and failure to thrive. The use of IBAT inhibitors has been shown to be effective in treating chronic diarrhea and poor growth in patients with FIC1 disease.

Long-Term Results of Pediatric Liver Transplantation for Progressive Familial Intrahepatic Cholestasis

Chenyue Hang et al.

Summary: This study analyzed the long-term survival rate and development of patients with progressive familial intrahepatic cholestasis (PFIC) after liver transplantation (LT). The results showed that LT is an effective treatment for end-stage PFIC patients with encouraging long-term survival rate and development. However, allograft steatosis should be closely monitored in PFIC type I patients.

A Report of 2 Infant Siblings with Progressive Intrahepatic Familial Cholestasis Type 1 and a Novel Homozygous Mutation in the ATP8B1 Gene Treated with Partial External Biliary Diversion and Liver Transplant

Irena Jankowska et al.

Summary: In patients with PFIC-1, the effect of PEBD may be delayed, even in cases of severe genetic mutations, emphasizing the need for cautious decision-making regarding the timing of LTx. Total biliary diversion is an effective procedure for persistent symptoms post LTx, and can reverse graft steatosis in children with PFIC-1.

Impact of Genotype, Serum Bile Acids, and Surgical Biliary Diversion on Native Liver Survival in FIC1 Deficiency

Daan B. E. van Wessel et al.

Summary: This study utilized the largest genetically defined cohort of patients with FIC1 deficiency to date, revealing that less than half of patients reach adulthood with native liver function; the number of PPTMs did not associate with the natural history or prognosis of FIC1 deficiency; sBA concentrations provide limited prognostic information on long-term NLS.

Odevixibat: First Approval

Emma D. Deeks

Summary: Odevixibat, a small molecule inhibitor of the ileal bile acid transporter, has received approvals in the EU and USA for the treatment of PFIC and pruritus associated with PFIC. It is also in clinical development for other cholestatic diseases in various countries.

Long-term Outcomes of Living-donor Liver Transplantation for Progressive Familial Intrahepatic Cholestasis Type 1

Tatsuya Okamoto et al.

Summary: The study revealed a correlation between post-LT outcomes in PFIC-1 patients with ATP8B1 genotypes and extrahepatic manifestations, especially intestinal function. Further research into this correlation could help identify patients who will achieve favorable post-LT outcomes.
