Small-molecule fluorescent probes based on covalent assembly strategy for chemoselective bioimaging

Significant contributions have been made in both the application and mechanism of covalent assembly probes over the past decade. This review summarizes the recent development of covalent assembly probes, classifies them based on different analytes, and introduces their detection mechanism in detail.

A novel mitochondrial homocysteine/cysteine-selective fluorescent probe has been successfully prepared, showing mitochondria-targeting property and sensitive responses toward Hcy/Cys. This probe could help broaden knowledge about the effects of biothiols, especially Hcy/Cys, and provide information on biothiol-relevant redox status.

Summary: A dicyanoisophorone/acrylate-combined probe (DDP) was synthesized as a near-infrared fluorescent sensor for the rapid identification of Cys over Hcy and GSH in aqueous solution. DDP had a large Stokes shift and a detection limit for Cys lower than the intracellular concentration. The probe was cell membrane-permeable and successfully applied for detecting intracellular Cys in HeLa cells, with the detection mechanism determined by various methods including H-1 NMR titration, MS, and DFT calculations.
