High-Resolution Genetic Mapping Combined with Transcriptome Profiling Reveals That Both Target-Site Resistance and Increased Detoxification Confer Resistance to the Pyrethroid Bifenthrin in the Spider Mite Tetranychus urticae

Summary: The study found that Boest1 expression levels in Chinese chive pests are age-dependent, with higher transcription levels in the midgut and Malpighian tubule during larval stages. Exposure to malathion and bifenthrin increased the expression of Boest1 significantly. BoEST1 exhibited strong catalytic activity towards alpha-naphthyl acetate in vitro, which was inhibited by malathion and triphenyl phosphate.

Summary: This study found that resistance to the organotin acaricide FBO in the spider mite Tetranychus urticae is polygenic. Two QTLs on chromosome 1 were associated with FBO resistance, and several cytochrome P450 monooxygenase (CYP) genes, including CYP392E4, CYP392E6 and CYP392E11, were located near the second QTL. A unique V89A mutation in the subunit c of mitochondrial ATP-synthase, located near the first QTL, was found to be enriched in the resistant parents and selected populations. Marker-assisted introgression experiments demonstrated a significant effect of the V89A mutation on the toxicity of organotin acaricides.

Overexpression of PxαE14 Contributing to Detoxification of Multiple Insecticides in Plutella xylostella (L.)

Summary: In this study, a carboxylesterase gene, Px alpha E14, was found to be significantly overexpressed in a field-evolved multiresistant population of the diamondback moth, Plutella xylostella. Px alpha E14 was predominantly expressed in the midgut and malpighian tubule of larvae. Knockdown of Px alpha E14 increased the susceptibility of the larvae to multiple insecticides, while overexpression of Px alpha E14 in fruit flies increased their tolerance to these insecticides. Recombinant Px alpha E14 expressed in Escherichia coli exhibited metabolic activity against multiple insecticides. These findings contribute to our understanding of the molecular mechanisms underlying multiresistance in insect pests.

Characterization of two kdr mutations at predicted pyrethroid receptor site 2 in the sodium channels of Aedes aegypti and Nilaparvata lugens

Summary: This study characterized the functional properties of the V253F and T267A mutations in the Ae. aegypti and N. lugens sodium channels. The results showed that these mutations reduced channel sensitivity to pyrethroids and altered channel gating properties. Combining V253F or T267A with F1534C or M918T mutations resulted in higher levels of pyrethroid insensitivity.

Pesticide Resistance and Related Mutation Frequencies of Tetranychus urticae in Hainan, China

The results showed high resistance to abamectin and bifenthrin, while the populations were susceptible or had low resistance to bifenazate and B-azolemiteacrylic.
