Gemcitabine-Resistant Biomarkers in Bladder Cancer are Associated with Tumor-Immune Microenvironment

Yuxuan Song et al.

Summary: Through analyzing large-scale sequencing data, we identified CAV1, COL6A2, FABP4, FBLN1, PCOLCE, and CSPG4 as critical genes in gemcitabine-resistant bladder cancer (BCa). These genes regulate the infiltration of tumor-infiltrating immune cells and are significantly associated with overall survival in BCa. Additionally, microRNAs play important regulatory roles in gemcitabine resistance.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Calcium channel α2δ1 subunit is a functional marker and therapeutic target for tumor-initiating cells in non-small cell lung cancer

Yuanyuan Ma et al.

Summary: Research has identified a subpopulation of TICs in non-small cell lung carcinoma (NSCLC) that can be specifically defined by the voltage-gated calcium channel alpha 2 delta 1 subunit, demonstrating stem cell-like properties and resistance to conventional chemotherapy. This discovery suggests that targeting alpha 2 delta 1 to prevent calcium influx provides a novel strategy for targeted therapy against TICs in NSCLC.

Lysosomes and Cancer Progression: A Malignant Liaison

Eda R. Machado et al.

Summary: The lysosomal system plays a crucial role in cancer progression, impacting communication between tumor cells and their microenvironment through changes in nutrient sensing, positioning, exocytosis, and fusion. This system has emerged as a potential therapeutic target in cancer treatment due to its versatile activities and interactions within cells.

The AML microenvironment catalyzes a stepwise evolution to gilteritinib resistance

Sunil K. Joshi et al.

Summary: The study reveals that the bone marrow microenvironment protects residual AML cells and AML cells develop resistance mechanisms over time. By integrating multiple methods, early resistant cells were found to undergo metabolic reprogramming and depend on AURKB, while late resistant cells are characterized by expansion of pre-existing NRAS mutant subclones and metabolic reprogramming.

High expression of PSMC2 promotes gallbladder cancer through regulation of GNG4 and predicts poor prognosis

Dawei Zhu et al.

Summary: This study identified PSMC2 as a potential regulator in the development of GBC, with high PSMC2 expression correlated with more advanced tumor grade and poorer prognosis. PSMC2 was revealed as a tumor promotor in GBC development, regulating cell phenotypes of GBC cells through interaction with GNG4.

Cytosolic DNA sensing by cGAS: regulation, function, and human diseases

Le Yu et al.

Summary: cGAS is a major cytosolic DNA sensor that, when hyperactivated, contributes to autoimmune diseases but serves as an adjuvant for anticancer immune therapy; inactivation of cGAS signaling causes difficulty in sensing and clearing infections, as well as facilitating tumor immune evasion, therefore cGAS activation is tightly controlled.

Endoplasmic reticulum stress signals in the tumour and its microenvironment

Xi Chen et al.

Summary: This review explores how ER stress in the tumor microenvironment can influence the functions of cancer cells and immune cells, leading to vulnerabilities that could be targeted by emerging therapeutic strategies. The tightly regulated processes of protein handling, modification, and folding in the ER determine cell function and survival, and disruptions in ER homeostasis can result in a state of persistent ER stress with pro-tumoral effects in cancer cells and immunomodulatory effects in immune cells.
