Local fluid shear stress operates a molecular switch to drive fetal semilunar valve extension

Duc H. Pham et al.

Summary: The study found that the elongation of the fetal semilunar valve occurs through fibrosa-sided endocardial proliferation. Low shear stress induces canonical Wnt signaling in fetal valve endothelium, driving endocardial proliferation, while high shear stress activates Notch signaling to inhibit Wnt signaling. This novel mechanically regulated molecular switch explains how fluid shear stress guides the growth direction of valve endothelium.

NOTCH Signaling in Aortic Valve Development and Calcific Aortic Valve Disease

Yidong Wang et al.

Summary: NOTCH signaling regulates multiple biological processes involved in tissue morphogenesis and homeostasis. Mutations in NOTCH1 can cause congenital bicuspid aortic valve and calcific aortic valve disease, with genetic variants affecting other genes in the pathway also contributing to these diseases. The role of NOTCH signaling in the postnatal homeostasis of the aortic valve is crucial, as animal studies have provided detailed insights into the developmental etiology of congenital aortic valve defects.

Prognostic Utility of Circulating Growth Factors in Aortic Valve Stenosis: A Pilot Study

Juris Hofmanis et al.

Summary: The study suggests that GDF-15 may serve as a clinically relevant biomarker for risk stratification in AS patients, showing a strong relationship with AS severity. In contrast, VEGF-A, FGF-2, and FGF-21 levels were significantly higher in AS patients but had weaker associations with disease severity.

Angptl2 is a Marker of Cellular Senescence: The Physiological and Pathophysiological Impact of Angptl2-Related Senescence

Nathalie Thorin-Trescases et al.

Summary: Cellular senescence is a cell fate induced by DNA damage, leading to irreversible growth arrest to prevent further damage. Senescence is a cellular response to stressors, observed in aging, wound healing, and embryonic development. Senescent cells secrete a variety of molecules in the SASP, including inflammatory cytokines and growth factors, with implications in both physiological and pathological conditions.

Mechanosensitive Notch-Dll4 and Klf2-Wnt9 signaling pathways intersect in guiding valvulogenesis in zebrafish

Alessio Paolini et al.

Summary: In the zebrafish embryo, blood flow generates fluid shear stress on endocardial cells, activating Notch and Klf2 signaling pathways crucial for atrioventricular valvulogenesis. Lateral inhibition mediated by Notch selects Delta-like-4-positive endocardial cells to ingress into the cardiac jelly and form an abluminal cell population. Wnt9a signaling cascade, activated by blood flow through Erk5Klf2-Wnt9a pathway, drives Delta-like-4-positive cells towards either luminal or abluminal fates, ultimately sculpting cardiac valve leaflets.
