Computational Identification of Druggable Bioactive Compounds from Catharanthus roseus and Avicennia marina against Colorectal Cancer by Targeting Thymidylate Synthase

Summary: This study aimed to identify natural anticancer agents that can inhibit the activity of thymidylate synthase (TS) protein, a potential drug target for colorectal cancer (CRC). Through molecular docking, molecular dynamics simulation, and other approaches, several compounds with better binding affinity to TS protein were identified. The selected compounds showed good pharmacokinetics and low toxicity. The study concluded that phytoconstituents from Catharanthus roseus and Avicennia marina have promising anticancer activity against TS protein and could be further developed as anti-CRC drugs.

Focal adhesion kinase priming in pancreatic cancer, altering biomechanics to improve chemotherapy

Summary: The dense desmoplastic and fibrotic stroma in pancreatic ductal adenocarcinoma (PDAC) plays a role in disease progression and treatment response. Reciprocal interactions between the tumor and stroma through integrin-mediated signaling, particularly Focal Adhesion Kinase (FAK), are important in regulating stromal remodeling and tissue stiffness. Targeting the PDAC stroma, including transient stromal manipulation via short-term FAK inhibition, may enhance the vulnerability of PDAC cells to chemotherapy. This article highlights the significance of treatment timing and suggests further exploration of anti-fibrotic therapies for PDAC and other fibrotic diseases.

Summary: This study utilized a computer-aided drug design approach to screen potential antiviral compounds against White spot syndrome virus (WSSV) from Withania somnifera plants in Saudi Arabia. Three compounds were identified as promising inhibitors of the virus based on their docking scores and further evaluated using pharmacokinetics and toxicity analyses. Molecular dynamics simulations confirmed their structural stability in the binding sites of the target protein. These selected compounds hold potential as antiviral drugs to combat WSSV and bring advancements to the aquaculture sector.
