Synthesis of 4-hydroxy-L-proline derivatives as new non-classical inhibitors of human carbonic anhydrase II activity: an in vitro study

Leila Moghoufei et al.

Summary: Carbonic anhydrase isozymes play important roles in pathological and physiological processes. In this study, 4-hydroxy-L-proline derivatives were found to inhibit the enzyme activity of hCA II, with compounds 3b and 3c exhibiting the lowest IC50 and Kd values. Further testing revealed that these compounds competitively inhibited hCA II esterase activity in a linear manner and reduced the hydrophobicity of hCA II surface.

Development of Hydrogen Sulfide-Releasing Carbonic Anhydrases IX- and XII-Selective Inhibitors with Enhanced Antihyperalgesic Action in a Rat Model of Arthritis

Alessandro Bonardi et al.

Summary: An effective therapeutic approach for arthritis management is proposed based on the anti-inflammatory role of hydrogen sulfide (H2S) and the inhibition of carbonic anhydrases (CAs) IX and XII. The designed H2S donor-CA inhibitor hybrid derivatives demonstrated enhanced inhibition potency and isoform selectivity, leading to significant pain relief in a rat model of arthritis.

Hypoxia-regulated carbonic anhydrase IX (CAIX) protein is an independent prognostic indicator in triple negative breast cancer

Chong Hui Clara Ong et al.

Summary: This study aims to investigate the effects of the hypoxia-inducible marker carbonic anhydrase IX (CAIX) in triple-negative breast cancer (TNBC) and its correlation with clinicopathological parameters and survival outcomes. The results demonstrate that CAIX is a significant hypoxia-inducible molecular marker in TNBC and increased CAIX levels are associated with poor survival.
