Fragment-Based Drug Discovery against Mycobacteria: The Success and Challenges

Namdev S. Togre et al.

Summary: The emergence of drug-resistant mycobacteria is a global threat that requires the development of new potent anti-mycobacterial drugs. Fragment-based drug discovery (FBDD) has been recognized as a popular approach to identify potent fragment molecules using virtual, computational, and biophysical methods. FBDD overcomes the limitations of traditional methods and is important for combating NTM and Mtb infections.

Comparative Analysis of Pharmacodynamics in the C3HeB/FeJ Mouse Tuberculosis Model for DprE1 Inhibitors TBA-7371, PBTZ169, and OPC-167832

Gregory T. Robertson et al.

Summary: The study evaluated the efficacy of three DprE1 inhibitors in a mouse tuberculosis model, showing significant effects after 2 months of treatment. OPC-167832 demonstrated superior efficacy at low dose levels, possibly due to its low MIC, favorable distribution, and sustained retention in lesion tissue.

Species-Specific Urothelial Toxicity With an Anti-HIV Noncatalytic Site Integrase Inhibitor (NCINI) Is Related to Unusual pH-Dependent Physicochemical Changes

Ruth A. Roberts et al.

Summary: GS-9695 and GS-9822, the next-generation noncatalytic site integrase inhibitors, show significantly improved potency against human immunodeficiency virus compared with previous drugs, but development was halted due to bladder urothelium vacuolation observed in cynomolgus monkeys. The underlying cause remains unclear despite extensive investigations into physicochemical changes and structural analysis of the compounds, providing valuable insights for future research and development of NCINIs.
