Low-dose metformin targets the lysosomal AMPK pathway through PEN2

Teng Ma et al.

Summary: The molecular target of the antidiabetic medicine metformin is identified as PEN2, and the PEN2-ATP6AP1 axis activates the AMPK pathway, providing therapeutic effects without substantial adverse effects.

Dose-dependent accumulation of glucose in the intestinal wall and lumen induced by metformin as revealed by 18F-labelled fluorodeoxyglucose positron emission tomography-MRI

Jun Ito et al.

Summary: The study aimed to investigate the relationship between clinical variables and FDG accumulation in the intestines of individuals with type 2 diabetes receiving metformin treatment. Results showed that metformin dose was an independent determinant of FDG accumulation in the large intestine wall and lumen, correlated with blood glucose levels and estimated glomerular filtration rate. SUVmax for the small intestine was not correlated with these variables.

Inhibition of mitochondrial function by metformin increases glucose uptake, glycolysis and GDF-15 release from intestinal cells

Ming Yang et al.

Summary: Recent studies have found that the gastrointestinal tract is an important site of action for metformin, affecting glucose uptake, glycolysis, and GDF-15 secretion likely due to observed mitochondrial dysfunction. These findings help explain the effects of metformin on intestinal glucose utilization and food balance.
