Selective inhibition of activated protein C anticoagulant activity protects against hemophilic arthropathy in mice

Jhansi Magisetty et al.

Summary: Recurrent spontaneous or trauma-related bleeding into joints in hemophilia leads to a debilitating joint disease called hemophilic arthropathy (HA). The treatment of HA involves preventing joint bleeding through clotting factor replacement and, in severe cases, orthopedic surgery. Recent research has shown that the administration of a specific monoclonal antibody (mAb) that blocks the activity of activated protein C (APC) offers a therapeutic potential for reducing the severity of HA without compromising its cytoprotective signaling.

Summary: (super)FVa can prevent ATC and traumatic bleeding when administered prophylactically, and can stabilize bleeding and reverse abnormal hemostasis parameters when administered while ATC is in progress.

Blocking human protein C anticoagulant activity improves clotting defects of hemophilia mice expressing human protein C

Miao Jiang et al.

Summary: This study tested the therapeutic potential of a monoclonal antibody, HAPC1573, that selectively blocks the anticoagulant activity of human activated protein C (APC). The results showed that HAPC1573 improved bleeding symptoms, reduced mortality and joint swelling, and enhanced thrombin generation in hemophilia mice.

Transportation of Single-Domain Antibodies through the Blood-Brain Barrier

Eduardo Ruiz-Lopez et al.

Summary: Single-domain antibodies derived from Camelidae and cartilaginous fishes show potential for diagnosis and treatment of CNS pathologies. While the blood-brain barrier presents a challenge for their delivery, certain neurological diseases and brain pathologies with BBB leakiness may favor their uptake into the CNS.
