A bioactive compliant vascular graft modulates macrophage polarization and maintains patency with robust vascular remodeling

Summary: Conventional synthetic vascular grafts have high failure rates due to mismatched mechanical properties with native vessels and poor regenerative potential. In this study, we developed a new bioactive synthetic vascular graft with tailored pore sizes to promote pro-regenerative macrophage populations and improve vascular regeneration. The grafts showed similar mechanical properties to native blood vessels and maintained patency and vascular regeneration in animal models.

Masquelet technique: Effects of vancomycin concentration on quality of the induced membrane

Summary: The effects of PMMA spacers loaded with different concentrations of vancomycin on the capacity of proliferative, osteogenic, and angiogenic capacity of induced membranes were investigated in this study. The results showed that low concentrations of vancomycin did not interfere with the functionality of induced membranes and even promoted their capacity, while high concentrations of vancomycin had negative effects on the functionality of induced membranes.

Bone defect treatment: does the type and properties of the spacer affect the induction of Masquelet membrane? Evidence today

Summary: The Masquelet technique has high clinical success rates in treating traumatic bone loss. This systematic review aims to answer questions regarding the ideal properties of spacers and the optimal timing for the second-stage operation. The review found that smooth PMMA spacers loaded with low concentrations of antibiotics exhibited powerful osteogenic properties. Other materials such as polypropylene and calcium sulfate also showed good results. Additionally, membranes older than 8 weeks were found to have similar regenerative capacities to younger ones.

Macrophages Characterization in an Injured Bone Tissue

Summary: The use of biomaterials shows promise in promoting wound healing of bone tissue. The study identified M2-like macrophages as the dominant macrophage type in the damaged area of a rat model, with a smaller number of M1-like macrophages. High expression of IL-1 beta and IL-6 cytokines was also observed in the membrane region. By employing two hybridization techniques, M2b-like macrophages were identified for the first time in cryosections of non-decalcified bone. Microspectroscopical analysis was found to be essential for accurate macrophage characterization. The study also highlighted the limitations of immunolabeling and the potential of in situ HCR for in vivo macrophage characterization.
