Summary: This study comprehensively identified HLA and non-HLA genetic susceptibility to methazolamide-induced Stevens-Johnson syndrome (MTZ-SJS)/toxic epidermal necrolysis (TEN) in Han Chinese. The study confirmed the association of HLA-B*59:01 and identified HLA-B*55:02 as a novel risk allele for MTZ-SJS/TEN. Molecular docking analysis showed that MTZ binds more stably to the risk HLA proteins.

Single-cell TCR and RNA sequencing indicate that clonotype-specific TCRs can recognize HLA-B*13:01-DDS complex to trigger inflammatory cytokines associated with DDS-DIHS.
