Indisulam Reduces Viability and Regulates Apoptotic Gene Expression in Pediatric High-Grade Glioma Cells

Aqua-(2-formylbenzoato)triphenyltin(IV) induces cell cycle arrest and apoptosis in hypoxic triple negative breast cancer cells

Summary: In this study, the therapeutic efficacy and molecular mechanism of action of aqua-(2-for-mylbenzoato) triphenyltin (IV) compound (OTC) in the MDA-MB-231 cell line were investigated. The results showed that OTC had stronger cytotoxic effects on hypoxic cells and induced cell apoptosis. Moreover, OTC downregulated the expression of certain proteins related to tumor development under hypoxic conditions.

Intracellular pH-mediated induction of apoptosis in HeLa cells by a sulfonamide carbonic anhydrase inhibitor

Summary: This study investigates the effects of a synthetic CAIX inhibitor on HeLa cancer cells. The compound I was found to inhibit cell proliferation, induce apoptosis, and affect the expression of genes and proteins related to apoptotic pathways. CAIX inhibition also caused changes in pH balance, disruption in organelle integrity, and increased intracellular reactive oxygen levels. The findings suggest that CAIX inhibition has potential in cancer treatment, and compound I could be a promising therapeutic strategy for aggressive tumors.

Indisulam targets RNA splicing and metabolism to serve as a therapeutic strategy for high-risk neuroblastoma

Summary: This study demonstrates that high-risk neuroblastoma is sensitive to indisulam, a drug that targets both RNA splicing and cellular metabolism. The researchers show that indisulam induces the loss of RBM39, accumulation of splicing errors, and growth inhibition in neuroblastoma models. Analysis of RNA sequencing and metabolic profiling data reveals disruptions to cell cycle and metabolism in response to indisulam. The authors also demonstrate complete tumor regression without relapse in mice models treated with indisulam, confirming the therapeutic potential of targeting both metabolism and RNA splicing in high-risk neuroblastoma.
