Adipocyte-Specific Modulation of KLF14 Expression in Mice Leads to Sex-Dependent Impacts on Adiposity and Lipid Metabolism

Summary: Genome-wide association studies have found that single nucleotide polymorphisms on the KLF14 gene are associated with an increased risk for metabolic syndrome and type 2 diabetes, particularly in women. In mice, the deletion of Klf14 in adipocytes leads to increased fat mass in females and decreased fat mass in males. Additionally, increasing KLF14 abundance in female adipocytes may offer a novel treatment option for metabolic abnormalities associated with obesity and T2D.

Summary: Comprehensive omics-based comparative analyses revealed significant differences in gene expression, lipid, and metabolite profiles in tissue, ADSCs, and adipocytes from lipedema patients compared to non-affected controls. Functional assays demonstrated that dysregulated Bub1 signaling drives increased proliferation of lipedema ADSCs, suggesting a potential mechanism for enhanced adipogenesis in lipedema. Identification of signaling networks driving lipedema provides potential molecular targets, including Bub1, for novel lipedema therapeutics.

Lymphatic function and anatomy in early stages of lipedema

Summary: This study investigated the lymphatic vessel anatomy and function in early stages of lipedema using near-infrared fluorescence lymphatic imaging. The results showed that while lower extremity lymphatic vessels were dilated in lipedema patients, their propulsion rates exceeded those of the control group. In contrast, upper extremity lymphatics were normal and no dermal backflow was observed in patients with Stage I and II lipedema.

CRISPR-enhanced human adipocyte browning as cell therapy for metabolic disease

This approach presents a promising therapeutic strategy to enhance metabolic homeostasis without the need for immunogenic Cas9 or delivery vectors in the recipient.
