Sensory nerves promote corneal inflammation resolution via CGRP mediated transformation of macrophages to the M2 phenotype through the PI3K/AKT signaling pathway

Kelan Yuan et al.

Summary: This study investigated the role of corneal sensory nerves in Pseudomonas aeruginosa keratitis. The results showed that sensory neurons and macrophages have a synergistic effect in promoting calcitonin gene-related peptide (CGRP) release. CGRP inhibits corneal inflammation and promotes the transformation of macrophages to the M2 phenotype through the PI3K/AKT signaling pathway.

Summary: This review discusses recent trends in preclinical pain modelling and provides recommendations for experimental designs that may increase translational success. Best practices in pain behaviour testing have shifted over the past decade due to technological advancements and the emphasis on rigor and reproducibility. The discussion focuses on four fundamental decisions in pain behavior experiments: choice of subject, choice of assay, laboratory environment, and choice of outcome measures.

Macrophages transfer mitochondria to sensory neurons to resolve inflammatory pain

Michiel van der Vlist et al.

Summary: The resolution of inflammatory pain is actively controlled by macrophages through the transfer of mitochondria to sensory neurons. This process requires the expression of CD200R on macrophages and the non-canonical CD200R ligand iSec1 on sensory neurons.

Synchronized cluster firing, a distinct form of sensory neuron activation, drives spontaneous pain

Qin Zheng et al.

Summary: This article explores the mechanisms underlying spontaneous pain and identifies a unique form of abnormal spontaneous activity. The study shows that this activity is triggered by sympathetic nerves that grow into ganglia after injury and is correlated with spontaneous pain behaviors. The findings contribute to the development of new treatments for spontaneous pain.

Exploring the Correlation Between the Regulation of Macrophages by Regulatory T Cells and Peripheral Neuropathic Pain

Hongyu Chen et al.

Summary: This study investigates the role of regulatory T cells in the differentiation of macrophages on dorsal root ganglion (DRG) and explores the mechanism of nociceptive signals in the signal transfer station. The findings suggest that regulatory T cells reduce the ratio of M1/M2 macrophages and alleviate neuroma pain by regulating macrophage polarization. These findings provide key insights into developing new strategies to manage painful neuroma.

Polarization of macrophages in the trigeminal ganglion of rats with pulpitis

Lu Gao et al.

Summary: The study observed the polarization response and dynamic distribution of macrophages in the trigeminal ganglion (TG) during the development of dental pulp inflammation. The results showed that M2 macrophages increased after CFA-induced pulpitis, peaked at 2 weeks, and returned to normal levels after 6 weeks, while the trend of M1 macrophages was opposite to that of M2 macrophages. The data suggests that macrophage polarization in the TG may play a role in the neuroinflammation response induced by dental pulpitis.

Blockade of AIM2 inflammasome or α1-AR ameliorates IL-1β release and macrophage-mediated immunosuppression induced by CAR-T treatment

Dan Liu et al.

Summary: This study demonstrates that CAR-T therapy induces activation of the AIM2 inflammasome in macrophages, leading to the release of bioactive IL-1 beta. Additionally, the alpha 1-AR-mediated adrenergic signaling enhances the priming of AIM2 inflammasome, while tumor cell DNA release triggers inflammasome activation. The interaction between CAR-T cells and tumor cells causes a phenotypic switch in macrophages, inhibiting the cytotoxicity of CAR-T cells and T cell proliferation through upregulation of PD-L1 and IDO.

β2-Adrenergic Receptor Enhances the Alternatively Activated Macrophages and Promotes Biliary Injuries Caused by Helminth Infection

Stephane Koda et al.

Summary: This study reveals that beta 2-AR deficiency alleviates hepatobiliary damage in mice infected with C. sinensis by decreasing the activation and infiltration of M2 macrophages and reducing the production of type 2 cytokines associated with liver fibrosis. Furthermore, it shows that macrophages from Adrb2(-/-) mice exhibit decreased M2 markers and reduced ERK/mTORC1 phosphorylation compared to those from Adrb2(+/+), indicating the role of beta 2-AR in enhancing type 2 immune response through the ERK/mTORC1 signaling pathway in macrophages and their involvement in liver fibrosis.
