Mapping human haematopoietic stem cells from haemogenic endothelium to birth

Vincenzo Calvanese et al.

Summary: The ontogeny of human haematopoietic stem cells (HSCs) involves distinct stages of differentiation at different haematopoietic sites, with unique transcription factor machinery and surface markers evolving throughout development. The transition of HSCs to the liver is marked by molecular shifts, including acquisition of maturity markers and suppression of surface antigens reflecting nascent HSC identity. HSC origin can be traced back to haemogenic endothelial cells, providing insight into the maturation process of functional HSCs.

Single cell analyses identify a highly regenerative and homogenous human CD34+hematopoietic stem cell population

Fernando Anjos-Afonso et al.

Summary: This study reveals an uncharacterized population of human CD34(+) HSCs, referred to as EPCR+ HSCs, which exhibit high repopulation and self-renewal abilities. Their unique transcriptomic wiring, associated with gene modules related to differentiated cell lineages, allows them to exhibit multilineage lineage output.

Bmi1 suppresses protein synthesis and promotes proteostasis in hematopoietic stem cells

Rebecca J. Burgess et al.

Summary: The study showed that deficiency of Bmi1 leads to increased division of HSCs without inducing senescence. Bmi1 promotes HSC quiescence by negatively regulating ARX expression and promoting proteostasis by suppressing protein synthesis.

Integrative Single-Cell RNA-Seq and ATAC-Seq Analysis of Human Developmental Hematopoiesis

Anna Maria Ranzoni et al.

Summary: Using single-cell RNA sequencing and single-cell assay for transposase-accessible chromatin sequencing, researchers identified three highly proliferative oligopotent progenitor populations downstream of hematopoietic stem cells (HSCs)/multipotent progenitors (MPPs) in human fetal liver and bone marrow. They observed dynamic changes in chromatin accessibility and lineage-specific transcription factor activity along the differentiation trajectory, providing a useful framework for future investigation of human developmental hematopoiesis.

HectD1 controls hematopoietic stem cell regeneration by coordinating ribosome assembly and protein synthesis

Kaosheng Lv et al.

Summary: This study reveals the role of the E3 ubiquitin ligase HectD1 in regulating HSC function through ribosome assembly and protein translation. Deficiency of HectD1 leads to accumulation of ZNF622 and eIF6 on 60S, resulting in 60S/40S joining defects. Depletion of Znf622 in Hectdl-deficient HSCs restores ribosomal subunit joining and protein synthesis, highlighting the importance of ubiquitin-coordinated ribosome assembly in HSC regeneration.

JAK-STAT in Early Hematopoiesis and Leukemia

Eirini Sofia Fasouli et al.

Summary: Hematopoietic stem cells (HSCs) are regulated by both extracellular signals from the bone marrow niche and intrinsic cell signals, including the critical JAK-STAT pathway. Mutations in this pathway are associated with defects in HSCs and hematologic malignancies, making it a potential target for leukemia therapy. Therapeutic targeting of JAK-STAT shows promise in treating leukemia based on the role of this pathway in HSC regulation.

Niches that regulate stem cells and hematopoiesis in adult bone marrow

Stefano Comazzetto et al.

Summary: This article presents an overview of the positioning of HSCs in the bone marrow and how their function is maintained, as well as discusses the regulatory role of niches in HSC function and hematopoiesis. Some important unresolved questions in this area have also been identified. The niches play a crucial role in maintaining HSC function and hematopoiesis.

Translational control of stem cell function

James A. Saba et al.

Summary: Stem cells are characterized by their ability to self-renew and differentiate into different cell types, and recent studies have shown that their behavior is closely tied to the regulation of protein synthesis by the ribosome. Understanding how translation mechanisms control stem cell function, including pluripotency and differentiation, may provide insights into mechanisms of tumorigenesis.
