Carboxymethyl chitosan microspheres loaded hyaluronic acid/gelatin hydrogels for controlled drug delivery and the treatment of inflammatory bowel disease

Summary: A pH-sensitive composite hyaluronic acid/gelatin hydrogel drug delivery system containing carboxymethyl chitosan microspheres loaded with CUR was fabricated for the treatment of IBD. In vitro studies showed good sustained release properties, with a drug release rate of 65% over 50 hours. In vivo pharmacokinetic experiments demonstrated that high levels of CUR were maintained in colon tissue for over 24 hours, exhibiting anti-inflammatory effects and inhibiting pro-inflammatory cytokines secretion. The formulation group of CUR@gels showed the best therapeutic effect on colitis in mice, indicating the potential of the composite gel delivery system for effective CUR delivery in colitis treatment.

MSCHS displayed enhanced survival capacity and better therapeutic outcomes compared to 3D-cultured MSCs (MSC3D), partially due to their superior PGE2 secretion in promoting tissue repair.

Summary: This study demonstrated that AB23A intervention effectively prevented colitis-associated cancer in male mouse models by reducing body weight loss, disease activity index, colon tumor load, tissue injury, and inflammatory cytokine changes. AB23A also decreased the activation of TLR, NF-kappa B, and MAPK pathways, while increasing the expression of mucin-2 and tight junction proteins. Additionally, AB23A interference led to alterations in gut microbiota composition with reduced levels of pathogenic bacteria and increased levels of beneficial bacteria. These findings suggest that AB23A has potential for future use in treating colitis-associated cancer.
