Effects of biophysical cues of 3D hydrogels on mesenchymal stem cells differentiation

Yutong Guo et al.

Summary: In 3D hydrogels, MSCs can actively remodel the surrounding matrix through proteolytic degradation and cell-exerted force, resembling the in vivo situation. Factors affecting hydrogel modifiability like matrix viscoelasticity and degradability regulate adhesion, morphology, and fate decision of MSCs. MSCs in 3D hydrogels utilize multiple mechanotransduction mechanisms, including integrin-actomyosin cytoskeleton system, ion channels, microtubule cytoskeleton, and self-secreted proteinaceous matrix.

PKM2-dependent metabolic skewing of hepatic Th17 cells regulates pathogenesis of non-alcoholic fatty liver disease

Maria E. Moreno-Fernandez et al.

Summary: Recent evidence suggests that Th17 cells play a key role in the pathogenesis of NAFLD, with a distinct population of inflammatory hepatic CXCR3(+)Th17 cells exacerbating the disease. The pathogenic characteristics of these cells include increased chromatin accessibility, glycolytic output, and production of IL-17A, IFN-γ, and TNF-α. Modulation of glycolysis pathways can potentially reverse the inflammatory vigor of ihTh17 cells and severity of NAFLD.

Cytoskeletal prestress: The cellular hallmark in mechanobiology and mechanomedicine

Farhan Chowdhury et al.

Summary: The cytoskeletal prestress plays a central role in mechanotransduction and mechanobiology, influencing cell stiffness, softening, and various cell functions. The evidence suggests that cytoskeletal prestress is a governing principle and a cellular hallmark in mechanobiology.

Adhesion-mediated mechanosignaling forces mitohormesis

Kevin M. Tharp et al.

Summary: Mitochondria control cell fate by producing energy and signals, and the physical characteristics of the extracellular matrix can also affect mitochondrial function. Cells can sense the physical properties of the extracellular matrix and activate a mitochondrial stress response to adaptively tune mitochondrial function. Adhesion-mediated mechanosignaling may play a role in altered mitochondrial functions observed in aging and cancer.
