Mesothelial cell-derived antigen-presenting cancer-associated fibroblasts induce expansion of regulatory T cells in pancreatic cancer

Huocong Huang et al.

Summary: Recent studies have identified a unique population of cancer-associated fibroblasts (apCAFs) derived from mesothelial cells, which play a critical role in pancreatic cancer progression. These apCAFs directly induce naive CD4(+) T cells to differentiate into regulatory T cells (Tregs) in an antigen-specific manner.

Glutamine-Directed Migration of Cancer-Activated Fibroblasts Facilitates Epithelial Tumor Invasion

Aida Mestre-Farrera et al.

Summary: Our study reveals that cancer-associated fibroblasts display a higher dependency on glutamine than epithelial tumor cells, leading to increased sensitivity to glutaminase inhibition. Furthermore, our results demonstrate that glutamine deprivation promotes CAF migration and invasion, facilitating the movement of tumor epithelial cells towards nutrient-rich territories. Cancer-associated fibroblasts induce monocytic myeloid-derived suppressor cell generation via IL-6/exosomal miR-21-activated STAT3 signaling to promote cisplatin resistance in esophageal squamous cell carcinoma

Summary: This study identified the correlation between monocytic myeloid-derived suppressor cells (M-MDSCs) and cisplatin resistance in patients with esophageal squamous cell carcinoma (ESCC). Furthermore, it was found that cancer-associated fibroblasts (CAFs) promoted the differentiation of monocytes into M-MDSCs through the secretion of interleukin-6 (IL-6) and exosome-packed microRNA-21 (miR-21), activating the STAT3 signaling pathway. The study highlights the potential of targeting the STAT3 signaling pathway as a strategy to reverse drug resistance in ESCC patients with high infiltration of CAFs and myeloid cells.

MicroRNA-370-3p shuttled by breast cancer cell-derived extracellular vesicles induces fibroblast activation through the CYLD/Nf-κB axis to promote breast cancer progression

Summary: EVs secreted by breast cancer cells can carry miR-370-3p to aggravate breast cancer by downregulating CYLD expression and activating the NF-kappa B signaling pathway. Breast cancer cell-derived EVs promote NF activation, whereas activated fibroblasts contribute to enhanced stemness, migration, invasion, and EMT of cancerous cells. Cancer-Associated Fibroblast-Mediated Cellular Crosstalk Supports Hepatocellular Carcinoma Progression

Mengjia Song et al.

Summary: This study revealed a cytokine-mediated cellular crosstalk and clinical network involving the CLCF1-CXCL6/TGF-beta axis, which regulates the positive feedback loop among CAFs, tumor stemness, and TANs, HCC progression, and patient prognosis. The up-regulation of the CLCF1-CXCL6/TGF-beta axis in clinical samples showed a correlation with increased cancer stem cells, N2-polarized TANs, tumor stage, and poor prognosis. This suggests the CLCF1 cascade could be a potential prognostic biomarker and targeting CLCF1/ ciliary neurotrophic factor receptor or ERK1/2 signaling could provide an effective therapeutic target for patients with HCC.

Promotion of cholangiocarcinoma growth by diverse cancer-associated fibroblast subpopulations

Summary: The study reveals that hepatic stellate cells (HSC) are the main source of cancer-associated fibroblasts (CAF), and HSC-derived CAF interact predominantly with tumor cells. In mouse models, CAF promote the progression of intrahepatic cholangiocarcinoma (ICC), while in patients, a high pan-CAF signature is associated with decreased survival and increased recurrence. Single-cell RNA sequencing segregates CAF into distinct subpopulations, with different interactions with tumor cells and promoting ICC growth.

Small extracellular vesicles containing miR-192/215 mediate hypoxia-induced cancer-associated fibroblast development in head and neck squamous cell carcinoma

Guiquan Zhu et al.

Summary: This study revealed the mechanisms by which hypoxic cancer cells induce CAF-like differentiation through secreting sEVs and TGF-beta, with miRNAs delivered by sEVs playing a crucial role in this process. Elevated levels of miR-192/215 in HNSCC tissue-derived sEVs indicated a hypoxic and aggressive cancer stroma, and miR-215 in tumor tissue-derived sEVs was associated with poor overall survival in HNSCC patients. The findings suggest that sEVs serve as a messenger to transfer miRNAs from cancer cells to fibroblasts, contributing to the remodeling of the hypoxic tumor microenvironment and indicating the potential of cancer tissue-derived sEVs as biomarkers.

The cross-talk between tumor cells and activated fibroblasts mediated by lactate/BDNF/TrkB signaling promotes acquired resistance to anlotinib in human gastric cancer

Zhijian Jin et al.

Summary: In this study, anlotinib inhibited gastric cancer (GC) cell growth by inducing apoptosis and G2/M phase arrest in a dose- and time-dependent manner. The interaction between epithelial and stromal cells in the tumor microenvironment affects the response of GC cells to anlotinib. Secreted lactate from GC cells activates cancer-associated fibroblasts (CAFs) to produce BDNF in a NF-kappa B-dependent manner, reducing the sensitivity of GC cells to anlotinib. Targeting the BDNF-TrkB pathway increased the sensitivity of GC cells to anlotinib in a human patient-derived organoid model.

miR-200 deficiency promotes lung cancer metastasis by activating Notch signaling in cancer-associated fibroblasts

Summary: miR-200 miRNAs have been identified as potent suppressors of lung adenocarcinoma metastasis, with their expression specifically repressed in metastatic lung adenocarcinomas and their decrease strongly correlating with poor patient survival. miR-200 deficiency promotes the proliferation and activation of adjacent cancer-associated fibroblasts, which in turn increases the metastatic potential of lung cancer cells. The functional interaction between cancer cells and CAFs, regulated by miR-200 targeting Notch ligands, constitutes an essential mechanism to promote metastatic potential.

CCM3 is a gatekeeper in focal adhesions regulating mechanotransduction and YAP/TAZ signalling

Shan Wang et al.

Summary: Wang et al. have identified CCM3 as a negative regulator of YAP/TAZ activation and mechanotransduction in focal adhesions, with crucial roles in controlling mesenchymal/stromal stem cell differentiation and metastasis. CCM3 functions as a gatekeeper in focal adhesions, regulating mechanotransduction and YAP/TAZ signaling by competing with focal adhesion kinase for binding to paxillin. Loss of CCM3 in cancer-associated fibroblasts leads to exacerbated tissue remodelling and metastasis dissemination in mouse models of breast cancer.
