Animal models for studies of alcohol effects on the trajectory of age- related cognitive decline

Summary: There is increasing interest in studying the interaction between ethanol use and aging on brain and cognition. Animal models are utilized to investigate the mechanisms underlying brain aging and cognitive decline related to age-related neurodegenerative diseases. However, most studies on elderly problems and diseases are conducted on healthy young animals, lacking clinical relevance. The validity of animal models is discussed, emphasizing age-related differences in anxiety, sensory-motor function, and procedural learning as confounding factors for the translation of preclinical results to clinical settings. The effects of ethanol on brain aging depend on dosage, acute or chronic treatment, withdrawal, and age examined. Important questions concern the dose and duration of ethanol treatment and their impacts on cognitive function trajectory due to lifelong alcohol use by humans. Key research areas include the specificity of alcohol effects on cognitive functions and brain regions that decline with age, as well as the interaction between alcohol and mechanisms or biomarkers of brain aging. Alternatively, moderate alcohol use may serve as a source of reserve and resilience against brain aging. Longitudinal studies are advantageous in detecting the effects of treatment on the emergence of cognitive impairment in middle age and reducing stress/anxiety associated with alcohol exposure and behavioral testing, which disproportionately affect aged animals. Lastly, the effects of alcohol on senescent neuro-physiology and biomarkers of brain aging are examined, especially the interaction between age and alcohol effects on inflammation, oxidative stress, N-methyl-D-aspartate receptor function, and excitatory/inhibitory synaptic transmission balance.
