Functionalized quinoxalinones as privileged structures with broad- ranging pharmacological activities

Summary: Quinoxalinones are a class of organic compounds that have attracted extensive attention for their potential applications. Despite the existence of synthetic strategies, there are limited reports on the pharmacological activities of quinoxalinone scaffolds from a medicinal chemistry perspective. However, multifunctional quinoxalinones, as privileged structures, possess various biological activities, including anticancer, neuroprotective, antibacterial, and more. This review helps facilitate the development of quinoxalinone derivatives in medicinal chemistry.

Photoinduced Three-Component Difluoroalkylation of Quinoxalinones with Alkenes via Difluoroiodane(III) Reagents

Summary: An environmentally friendly strategy for a photo- catalyzed three-component reaction between quinoxalinones, alkenes, and hypervalent iodine(III) reagents is disclosed. The designed difluoroiodane(III) reagent shows excellent reactivity and can provide a wide range of difluoroalkyl-substituted quinoxaline-2(1H)-ones in moderate to excellent yields under mild conditions. Experimental studies confirm the involvement of a difluoroalkyl radical intermediate in the reaction.

Summary: In this study, the palladium-catalyzed asymmetric acyl-carbamoylation reaction was reported, producing alpha-quaternary chiral cycloketone with high yields and excellent enantioselectivity. The reaction proceeded via asymmetric 1,2-migratory insertions of acyl-Pd into alkenes and subsequent migratory insertion of isocyanides into C(sp(3))-Pd-II. The product could be further diversified, showcasing the synthetic utility of this protocol.
