Tumor-Microenvironment-Activated Reactive Oxygen Species Amplifier for Enzymatic Cascade Cancer Starvation/Chemodynamic /Immunotherapy

Man Wang et al.

Summary: A TME-responsive nanosystem was engineered in this study to enhance reactive oxygen species production in the tumor microenvironment for cancer starvation, chemodynamic, and immunotherapy, which was achieved through combining natural enzymes and nanozymes. Furthermore, the enhanced oxidative stress by the nanosystem facilitated antitumor immune responses and effectively inhibited tumor growth and metastasis when combined with PD-L1 antibody for synergistic therapy.

Photoacoustic mediated multifunctional tumor antigen trapping nanoparticles inhibit the recurrence and metastasis of ovarian cancer by enhancing tumor immunogenicity

Xiaowen Zhong et al.

Summary: The hypoimmunogenicity of tumors is a major obstacle in cancer immunotherapy. This study developed multifunctional tumor antigen trapping nanoparticles that enhance tumor immunogenicity through photoacoustic/ultrasound imaging and therapy. The nanoparticles successfully induced tumor antigen exposure and release, and inhibited tumor growth and metastasis. This research provides a simple and effective strategy for the treatment of metastatic and recurrent tumors.

A novel afterglow nanoreporter for monitoring cancer therapy

Shiyi Liao et al.

Summary: A novel near-infrared organic afterglow nanoparticle was developed to visualize reactive oxygen species (ROS) during immunogenic cell death, with high signal to noise ratio. The nanoparticle induced immunogenic cell death of cancer cells and caused immune responses, providing a non-invasive tool for predicting the degree of immunogenic cell death during ROS-mediated cancer therapy.

Exosome-based photoacoustic imaging guided photodynamic and immunotherapy for the treatment of pancreatic cancer

Yongho Jang et al.

Summary: In this study, tumor-derived reassembled exosome (R-Exo) was used both as a drug delivery carrier and an immunostimulatory agent for cancer therapy. By loading chlorin e6 photosensitizer into R-Exo, it retained its original size and membrane proteins for targeting tumor cells. The modified exosomes were able to generate reactive oxygen species and increase cytokine release from immune cells, showing their potential as an immunotherapeutic agent.

Antimony Nanopolyhedrons with Tunable Localized Surface Plasmon Resonances for Highly Effective Photoacoustic-Imaging-Guided Synergistic Photothermal/Immunotherapy

Yu Chen et al.

Summary: This study presents an effective strategy to synthesize antimony nanopolyhedrons and successfully observes the modulation effect of different morphologies on their localized surface plasmon resonance in experimentation. Antimony nanopolyhedrons exhibit excellent performance in PTCE and good photothermal stability, as well as biocompatibility. The research also demonstrates the construction of multifunctional nanomedicines based on antimony nanopolyhedrons for efficient photoacoustic-imaging-guided synergistic photothermal/immune-therapy of tumors in vivo.

Macrophage Polarization States in the Tumor Microenvironment

Ava J. Boutilier et al.

Summary: The M1/M2 macrophage paradigm is crucial in tumor progression, where M1 macrophages have anti-tumor effects and M2 macrophages contribute to tumor development through various mechanisms. The tumor microenvironment plays a role in influencing macrophage polarization and activation, which is essential for understanding the pro-tumor effects of tumor-associated macrophages.

Cancer-cell-biomimetic Upconversion nanoparticles combining chemo-photodynamic therapy and CD73 blockade for metastatic triple-negative breast cancer

Feiyang Jin et al.

Summary: The combined application of anti-CD73 antibody and chemo-PDT can enhance the anti-metastatic effects of T cell-mediated antitumor immunity synergistically. This strategy not only prevents abscopal tumor metastasis but also induces a series of immunogenic cell death, leading to tumor-specific immunity.

Peiqi Zhao et al.

Summary: This study developed a combined photothermal immunotherapy strategy using BBPQDs for TNBC treatment, showing enhanced antitumor activity compared to anti-PD-L1 monotherapy. The strategy effectively inhibits tumor recurrence and metastasis by reprogramming the tumor microenvironment and inducing immune memory against tumor rechallenge.
