WRKY33-mediated indolic glucosinolate metabolic pathway confers resistance against Alternaria brassicicola in Arabidopsis and Brassica crops

Han Tao et al.

Summary: This study reveals differential regulation of the tryptophan-derived camalexin and indolic glucosinolate (IGS) biosynthetic pathways in plant immunity between Arabidopsis and Brassica crops. The loss of camalexin biosynthesis in Brassica crops during evolution may attenuate their resistance to Alternaria brassicicola, highlighting the importance of the IGS metabolic pathway mediated by WRKY33 in Chinese kale's defense against Alternaria brassicicola.

Summary: This study revealed the genetic structure and breeding history of low seed glucosinolates (SGC) improvement in rapeseed by GWAS and TWAS, identified 15 reliable QTLs associated with reduced SGC in modern cultivars, and highlighted the candidate gene BnaC02.GTR2. Experimental validation showed that BnaC02.GTR2 and its paralogs positively regulate seed glucosinolate accumulation, with implications for seed development and potential for crop improvement.

Structural Studies of Aliphatic Glucosinolate Chain-Elongation Enzymes

Vivian Kitainda et al.

Summary: Plants have evolved specialized metabolic pathways through gene duplication and functional divergence, resulting in a variety of natural products beneficial to plants and humans. Glucosinolates, a diverse class of natural products in plants, have functions such as providing strong flavors in Brassicales plants, acting as plant defense molecules, and potentially serving as cancer prevention agents in humans. The structural diversity of glucosinolates allows for their wide range of functions, with aliphatic methionine-derived glucosinolates being the most abundant form. The mechanisms of elongation and chemical modification of amino acid side chains in glucosinolates, particularly in aliphatic glucosinolates, are still not fully understood at the molecular level.

Metabolite analysis of Arabidopsis CYP79A2 overexpression lines reveals turnover of benzyl glucosinolate and an additive effect of different aldoximes on phenylpropanoid repression

Veronica C. Perez et al.

Summary: The overexpression of the PAOx-producing enzyme in Arabidopsis leads to the accumulation of PAOx-derived compounds as well as the metabolism of benzyl glucosinolate, indicating a potential turnover process that does not involve known enzymes. The expression of myrosinases or beta-glucosidases did not substantially increase in the overexpressing lines, suggesting alternative pathways for the breakdown of glucosinolates.
