New Insights into the Mechanism of Antibacterial Action of Synthetic Peptide Mo-CBP3-PepI against Klebsiella pneumoniae

Levi A. C. Branco et al.

Summary: Fluorescence microscopy and proteomic analysis revealed that Mo-CBP3-PepI alters cell membrane permeability, induces ROS overproduction, and affects protein profile of K. pneumoniae cells. The proteomic analysis also highlighted changes in essential metabolic pathways. Mo-CBP3-PepI has potential as an alternative molecule to combat K. pneumoniae infections.

Membrane Integrity Contributes to Resistance of Cryptococcus neoformans to the Cell Wall Inhibitor Caspofungin

Brenda Moreira-Walsh et al.

Summary: The intrinsic resistance of Cryptococcus neoformans to caspofungin limits the available therapies for treating cryptococcal infections. By screening gene deletion strains, researchers identified biological processes associated with altered membrane permeability that could render the cell more susceptible to caspofungin. Targeting these genes or other genes affecting membrane permeability may provide potential for novel therapies against this global fungal pathogen.

Antifungal Potential of Synthetic Peptides against Cryptococcus neoformans: Mechanism of Action Studies Reveal Synthetic Peptides Induce Membrane-Pore Formation, DNA Degradation, and Apoptosis

Tawanny K. B. Aguiar et al.

Summary: In this study, synthetic antimicrobial peptides (SAMPs) were found to have antifungal effects on Cryptococcus neoformans. The SAMPs inhibited fungal growth by damaging the cell membrane and inducing DNA degradation. These findings suggest that SAMPs could be potential alternative molecules for controlling C. neoformans growth and treating cryptococcal infection.

Mechanisms of Action of the Antimicrobial Peptide Cecropin in the Killing of Candida albicans

Cui Peng et al.

Summary: The development of drug resistance has made fungal infections a global health concern. Antimicrobial peptides (AMPs) provide a viable solution to these pathogens due to their resistance to drug resistance and diverse mechanisms of action. This study identifies the genetically engineered peptide Cecropin as having antifungal effects against Candida albicans through mechanisms such as changing the permeability and fluidity of the cell membrane and inducing the production of reactive oxygen species.

Synthetic peptides against Trichophyton mentagrophytes and T. rubrum: Mechanisms of action and efficiency compared to griseofulvin and itraconazole

Patricia G. Lima et al.

Summary: The study found that the AMPs tested were more effective in inhibiting the growth of T. mentagrophytes and T. rubrum compared to griseofulvin and itraconazole. AMPs caused severe damage to hyphal morphology by inducing cell wall rupture, hyphal content leakage, and cell death, which was not observed with the commercial drugs. These findings suggest that the synthetic peptides have the potential to be developed as new antidermatophytic drugs or used as adjuvants to existing ones.

Advances in Antifungal Drug Development: An Up-To-Date Mini Review

Ghada Bouz et al.

Summary: The clinical utility of antifungal drugs is hindered by their limited scope of activity, high toxicity, and emerging resistance. Current research is focused on developing novel antifungal agents, including first-in-class drugs, new structures for established targets, and modifications to existing antifungal formulations.
