Synergetic Fermentation of Glucose and Glycerol for High-Yield N-Acetylglucosamine Production in Escherichia coli

Kaikai Wang et al.

Summary: This study presents a synergistic substrate co-utilization strategy to improve the production efficiency of GlcNAc by adjusting carbon flux distribution. By blocking the main glucose catabolism pathways and introducing the GlcNAc production module, high stoichiometric yield of GlcNAc was achieved.

Exploring the Cystic Fibrosis Lung Microbiome: Making the Most of a Sticky Situation

Christina S. Thornton et al.

Summary: Chronic lower respiratory tract infections are a major contributor to morbidity and mortality in patients with cystic fibrosis. The microbial community structure in the airways evolves throughout the lifetime of patients and is associated with baseline disease state, disease progression, and occurrence of pulmonary exacerbations. Molecular analysis of the airway microbiome provides valuable insights, but challenges remain in understanding the relationship between microbial presence and activity and disease progression.

Polyclonality, Shared Strains, and Convergent Evolution in Chronic Cystic Fibrosis Staphylococcus aureus Airway Infection

Dustin R. Long et al.

Summary: This study characterized the molecular epidemiology and genetic adaptation of Staphylococcus aureus in chronic CF airway infection, revealing that patients often carry multiple, genetically distinct S. aureus lineages with adaptations in persistence and antimicrobial resistance genes. Limited strain-sharing was identified among patients, and S. aureus genetic diversity was found to be unconstrained, suggesting the need for novel diagnostic and therapeutic approaches in the future.

Inflammation in children with cystic fibrosis: contribution of bacterial production of long-chain fatty acids

Erin Felton et al.

Summary: Long-chain fatty acid biosynthesis pathways were upregulated in follow-up samples compared to the end of treatment, and Achromobacter xylosoxidans and Escherichia sp. were more prevalent in follow-up. LCFAs may be associated with persistent infection of opportunistic pathogens. Future studies should investigate the role of bacterial production of LCFAs in CF patients' transition from baseline wellness to PEx.

How Bacterial Adaptation to Cystic Fibrosis Environment Shapes Interactions Between Pseudomonas aeruginosa and Staphylococcus aureus

Laura Camus et al.

Summary: Pseudomonas aeruginosa and Staphylococcus aureus are the two most prevalent bacteria species in the lungs of cystic fibrosis (CF) patients. Co-infection by the two species is common, and recent studies have shown that they are able to coexist, with this change in relationship linked to bacterial strain evolution.

An observational study of anaerobic bacteria in cystic fibrosis lung using culture dependant and independent approaches

Claudie Lamoureux et al.

Summary: This study described the diversity of anaerobic bacteria in the CF airway microbiota, identifying 31 anaerobic genera and 69 species. Better lung function was significantly associated with higher quantification of Veillonella, raising questions about the potential impact of anaerobes on lung function.

Integrating taxonomic, functional, and strain-level profiling of diverse microbial communities with bioBakery 3

Francesco Beghini et al.

Summary: Culture-independent analyses of microbial communities have advanced significantly in the past decade, particularly due to the emergence of Shotgun Metagenomics. bioBakery 3, a newly developed toolset, enhances the accuracy and scope of taxonomic, functional, and phylogenetic profiling in microbial community studies by providing an expanding set of reference sequences.

Importance of beta-lactam pharmacokinetics and pharmacodynamics on the recovery of microbial diversity in the airway of persons with cystic fibrosis

Andrea Hahn et al.

Summary: The study found that the pharmacokinetic target attainment of beta-lactam antibiotics in children with cystic fibrosis was associated with suppressed recovery of airway microbial diversity following treatment. Patients with sufficient antibiotic levels had decreased alpha diversity at follow-up, while those with insufficient levels showed an increase in diversity during the same period. Inadequate beta-lactam levels were also linked to a higher frequency of fluoroquinolone resistance.

Staphylococcus aureus and Cystic Fibrosis-A Close Relationship. What Can We Learn from Sequencing Studies?

Christine Rumpf et al.

Summary: Staphylococcus aureus is a common pathogen in the airways of CF patients, which adapts and evolves in this specific environment through horizontal gene transfer and nucleotide variations. Methicillin-resistant S. aureus poses a greater risk for infected individuals with more severe lung disease.
