Trimethylamine N-Oxide Levels in Non-Alcoholic Fatty Liver Disease: A Systematic Review and Meta-Analysis

Changes of flavin-containing monooxygenases and trimethylamine-N-oxide may be involved in the promotion of non-alcoholic fatty liver disease by intestinal microbiota metabolite trimethylamine

Summary: This study explored the role of trimethylamine (TMA) in nonalcoholic fatty liver disease (NAFLD) and found that TMA and its metabolite trimethylamine-N-oxide (TMAO) are closely related to NAFLD. The expression of flavin-containing monooxygenase 1 (FMO1) was increased in NAFLD, and FMO1 and its metabolite TMAO were induced by high fat and TMA. However, silencing FMO1 blocked the effects of high fat and TMA on TMAO. Furthermore, TMAO may exert its role in NAFLD through the activation of unfolded protein response (UPR).

Incidence of Hepatocellular Carcinoma in Patients With Nonalcoholic Fatty Liver Disease: A Systematic Review, Meta-analysis, and Meta-regression

Summary: Nonalcoholic fatty liver disease (NAFLD) may be a risk factor for hepatocellular carcinoma (HCC), and this study used meta-analysis to analyze the incidence rates of HCC in NAFLD patients. The results showed that NAFLD patients with cirrhosis have a similar risk of developing HCC compared to cirrhosis patients with other causes. However, the incidence of HCC in nonalcoholic steatohepatitis or simple steatosis patients is lower.

It is mainly derived from the gut where gut microbiota converts TMA precursors into TMA, which is then transformed into TMAO in the liver. High-nutrient diets rich in TMA precursors are the main sources of TMAO and can directly affect energy metabolism in the liver, promoting the development of MAFLD.
