MiR-181c suppresses triple-negative breast cancer tumorigenesis by targeting MAP4K4

Summary: Breast cancer is the most common cancer in women worldwide, and triple negative breast cancer is a highly aggressive subtype of breast cancer. This study demonstrates that miR-181c inhibits the proliferation and migration of triple negative breast cancer cells, promotes apoptosis, and regulates the cell cycle. Importantly, miR-181c suppresses the tumor-promoting effect of MAP4K4 by targeting its expression.

Promoting Apoptosis, a Promising Way to Treat Breast Cancer With Natural Products: A Comprehensive Review

Summary: Breast cancer is a major global health concern for women due to its high rates of morbidity and mortality. Chemotherapy is commonly used for breast cancer treatment, but resistance and toxic side effects are common. In recent years, natural products have gained attention for their low side effects, low toxicity, and effectiveness through multitarget therapy. Inducing apoptosis, a form of programmed cell death, is an important strategy for controlling excessive breast cancer cell proliferation. This review examines the current research on natural product-induced breast cancer cell apoptosis and summarizes the mechanisms involved. The hope is that this review will contribute to the search for candidate drugs derived from natural products to promote cell apoptosis and treat breast cancer.

Targeting HIF1-alpha/miR-326/ITGA5 axis potentiates chemotherapy response in triple-negative breast cancer

Summary: In this study, we identified fibronectin (FN1) as a central driver gene in chemoresistance. Overexpression of miR-326 reversed FN1-driven chemoresistance by targeting the FN1 receptor, ITGA5. miR-326 expression was downregulated by increased hypoxia/HIF1A and ECM stiffness in chemoresistant tumors, leading to upregulation of ITGA5 and activation of the downstream FAK/Src signaling pathways. Overexpression of miR-326 or inhibition of ITGA5 overcame FN1-driven chemotherapy resistance in vitro by inhibiting the FAK/Src pathway and potentiated the efficacy of chemotherapy in vivo. Importantly, lower expression of miR-326 or higher levels of predicted miR-326 target genes were significantly associated with worse overall survival in chemotherapy-treated TNBC patients.
