A Novel Real Time PCR Method for the Detection and Quantification of Didymella pinodella in Symptomatic and Asymptomatic Plant Hosts

Adnan Sisic et al.

Summary: A real-time PCR assay based on the TEF-1 alpha gene sequence alignments was developed for the detection and quantification of Didymella pinodella. The assay showed high specificity and consistency, with a quantification/detection limit of 3.1 pg of target DNA per reaction.

Molecular identification and pathogenicity of Rhizoctonia solani and Pythium spp. associated with damping-off disease on baby leafy vegetables in Greece

George T. Tziros et al.

Summary: In recent years, the cultivation of leafy vegetables as baby leaves has gained recognition in the global market and has attracted the interest of consumers. This study conducted a comprehensive analysis of specific Rhizoctonia solani AGs and Pythium spp. derived from baby leafy vegetables exhibiting damping-off symptoms in Greece. The results showed variations in pathogenicity among the different AGs of R. solani and identified three Pythium spp. The findings highlight the importance of understanding these pathogens in order to prevent and manage damping-off symptoms in baby leaf vegetables.

Detection and Quantification of Rhizoctonia solani and Rhizoctonia solani AG1-IB Causing the Bottom Rot of Lettuce in Tissues and Soils by Multiplex qPCR

Therese Wallon et al.

Summary: Vegetable growers in the muck soil region of southwestern Quebec are threatened by soilborne diseases like bottom rot caused by Rhizoctonia solani, with disease severity increasing due to warm temperatures. This study identified Rhizoctonia solani AG 1-IB as the main cause of bottom rot in field-grown lettuce and developed specific qPCR assays for detection. The assays showed sensitivity in detecting the pathogen in plant tissue and soil samples, providing useful tools for lettuce bottom rot management.

Application of Real-Time PCR for the Detection and Quantification of Oomycetes in Ornamental Nursery Stock

Alexandra Puertolas et al.

Summary: Numerous outbreaks of Phytophthora and Pythium diseases have occurred in Europe due to introduction of contaminated ornamental plants. Detection and identification of pathogens are crucial. Results showed that oomycetes were isolated from 89.9% of plants using classical techniques, and real-time PCR demonstrated that oomycetes were associated with all plants tested.
