Real-time detection of volatile metabolites enabling species-level discrimination of bacterial biofilms associated with wound infection

Elisabeth A. Slade et al.

Summary: This study successfully differentiated between different pathogens by real-time detection of volatile metabolites produced in a collagen wound biofilm model. The development of rapid point-of-care diagnostics for wound infection using this method could improve patient care and reduce antimicrobial resistance.

Practical challenges and potential approaches to predicting low-incidence diseases on farm using individual cow data: A clinical mastitis example

D. M. Liebe et al.

Summary: This study aimed to build models for predicting clinical mastitis (CM) incidence using time-series sensor data. The study found that model accuracy was crucial in terms of specificity (Sp) and positive predictive value. Additionally, there was an asymmetry between sensitivity (Se) and Sp when the disease incidence rate is low. Consequently, future CM predictive models should consider economic and practical implications.

Diversity and pathogenesis of Staphylococcus aureus from bovine mastitis: current understanding and future perspectives

Bruno Campos et al.

Summary: This review summarizes the research on bovine Staphylococcus aureus population genetics and molecular pathogenesis conducted worldwide in the past 16 years. It describes the diversity and geographical distribution of S. aureus associated with bovine mastitis, and explores the evolution of bovine S. aureus and the importance of host-adaptation in its emergence as a mastitis pathogen. The functional relevance of virulence determinants in the pathogenesis of bovine mastitis is also discussed.

Use of GC-IMS for detection of volatile organic compounds to identify mixed bacterial culture medium

Yanyi Lu et al.

Summary: This study explored the possibility of identifying common wound infection bacteria in mixed culture using gas chromatograph-ion migration spectroscopy (GC-IMS). The headspace gas of single and mixed cultures of Escherichia coli, Staphylococcus aureus, and Pseudomonas aeruginosa was analyzed, and specific volatile organic compounds (mVOCs) were obtained. The findings suggest that GC-IMS can be used to identify compound bacteria based on specific mVOCs.

Antimicrobial Resistance and Molecular Characterization of Staphylococcus aureus Recovered from Cows with Clinical Mastitis in Dairy Herds from Southeastern Brazil

Gustavo Freu et al.

Summary: Staphylococcus aureus is a contagious pathogen associated with bovine mastitis in Brazil. Molecular characterization of Staph. aureus isolates from affected cows provided data on the epidemiology and genetic-level antimicrobial susceptibility (AMS). The study found high genotypic similarity among Staph. aureus isolates within and between herds, with resistance to most antibiotics, particularly beta-lactam antibiotics.

Susceptibility Testing by Volatile Organic Compound Detection Direct from Positive Blood Cultures: A Proof-of-Principle Laboratory Study

Sacha Danielle Kuil et al.

Summary: Bacteria produce VOCs during growth, which can be detected by CSAs. The SpecifAST (R) system enables AST directly from blood cultures, with results available within 3.1 hours of growth detection without subculture steps. In a 12-month study, the system showed 100% and 91% categorical agreement with VITEK (R) 2 for Gram-negatives and Gram-positives, respectively. Further evaluation with a larger sample of bacterial species and antimicrobials is needed.

Summary: The study found that a small number of S. aureus ST types (ST352, ST2187, ST3028, and ST151) are associated with the majority of cases of bovine mastitis in Pennsylvania dairy farms. Additionally, it was observed that one ST of S. aureus predominated in the herd and this ST can coexist with several other ST types of S. aureus strains.

Summary: The detection of chemical compounds in exhaled breath presents an opportunity to determine physiological state, diagnose disease or assess environmental exposure. Despite some challenges, exhaled breath is desirable for metabolomics applications due to its non-invasiveness, convenience, and practical availability. Tests targeting endogenous or exogenous gas-phase compounds in breath are currently established for practical and clinical use.
