Unraveling Capsule Biosynthesis and Signaling Networks in Cryptococcus neoformans

Summary: IRIS is an exaggerated immune reaction that occurs in immunocompromised patients with late-stage HIV infection who have started ART, caused by virtually any opportunistic pathogen. Recent research focuses on identifying risk factors for Cryptococcal IRIS and understanding C-IRIS immunopathogenesis, with new findings in blood and cerebrospinal fluid potentially useful in predicting and diagnosing CM-IRIS. Current therapeutic regimens and novel treatment approaches for combating CM-IRIS are also discussed, along with the utility of biomarkers for clinical monitoring and adjusting treatment modalities in AIDS patients co-infected with Cryptococcus who have initiated ART.

Quantitative analysis reveals internalisation of Cryptococcus neoformans by brain endothelial cells in vivo

Summary: Migration of Cryptococcus neoformans from the blood to the brain parenchyma is crucial in causing fatal meningoencephalitis. A novel approach using flow cytometry in a mouse model was used to quantitatively analyze the interactions between C. neoformans and brain endothelial cells. Results showed that brain endothelial cells internalized C. neoformans in vivo and that different strains of the fungus had varying rates of invasion. Overall, the study provided a powerful method to quantitatively analyze fungal migration into the brain.
