RamAp Is an Efflux Pump Regulator Carried by an IncHI2 Plasmid

Yu-Ping Hong et al.

Summary: The investigation into the epidemiological trends of Salmonella enterica serovar Goldcoast revealed closely related strains with varying drug resistances. The comparison of two multidrug-resistant strains led to the identification of an additional ramA gene that can be transmitted through plasmids among bacteria. Introducing this gene into Escherichia coli resulted in elevated expression of efflux pump genes and increased minimum inhibitory concentrations to multiple antibiotics.

Identification and genetic characterization of two conjugative plasmids that confer azithromycin resistance in Salmonella

Miaomiao Xie et al.

Summary: With the development of multidrug resistance in Salmonella spp., ciprofloxacin, ceftriaxone, and azithromycin have become the main drugs used to treat Salmonella infections. However, not much research has been focused on azithromycin resistance in Salmonella. This study investigated the genetic features of plasmids encoding azithromycin resistance in food-borne Salmonella strains. The study found that these plasmids might originate from Escherichia coli and could play a role in the rapid spread of azithromycin resistance in Salmonella. The study also observed the formation of a new plasmid that poses a serious health threat. Further research on the dissemination and evolution routes of these plasmids in potential human pathogens is necessary.

A nosocomial salmonellosis outbreak caused by blaOXA-48-carrying, extensively drug-resistant Salmonella enterica serovar Goldcoast in a hospital respiratory care ward in Taiwan

Chih-Ming Chen et al.

Summary: A nosocomial salmonellosis outbreak occurred in a respiratory care ward of a hospital in central Taiwan, with 10 isolates showing resistance to extended-spectrum cephalosporins. Whole-genome sequencing revealed that the outbreak strains carried multiple antimicrobial resistance genes and acquired a bla(OXA-48)-carrying plasmid, resulting in carbapenem resistance. The spread of extremely resistant strains should be closely monitored in Taiwan.

Multidrug Resistance Genes Carried by a Novel Transposon Tn7376 and a Genomic Island Named MMGI-4 in a Pathogenic Morganella morganii Isolate

Xing-Wei Luo et al.

Summary: Mobile genetic elements (MGEs), especially IS26-bounded translocatable units, can serve as a reservoir for a variety of antimicrobial resistance genes in clinically important pathogenic bacteria. This study investigated a representative M. morganii isolate containing multidrug resistance genes, including the infrequent dfrA24, to provide insights into this significant genetic characteristic. The acquired resistance genes in M. morganii were predominantly carried by important MGEs flanked by IS26, such as the novel Tn7376 and the MMGI-4. The emergence of the extra gene dfrA24 in M. morganii was likely facilitated by various genetic events, including recombination, transposition, and integration mediated by IS26-related MGEs. Therefore, continuous surveillance of these MGEs in clinical pathogens is crucial for assessing the risk of emergence and development of novel resistance genes.

Group Author Only

Summary: Data on antimicrobial resistance in humans, animals, and food are collected annually by EU Member States and jointly analysed by EFSA and ECDC. The 2020 monitoring focused on poultry and their meat, while the 2019 monitoring focused on fattening pigs, calves, and their meat. Monitoring included data on Salmonella, Campylobacter, E. coli isolates, and specific monitoring of ESBL-/AmpC-/carbapenemase-producing E. coli isolates. Some countries also reported data on methicillin-resistant Staphylococcus aureus.

Integrative and Conjugative Element-Mediated Azithromycin Resistance in Multidrug-Resistant Salmonella enterica Serovar Albany

Yu-Ping Hong et al.

Summary: In cases of human salmonellosis in Taiwan between 2014 and 2019, a study found that 26.4% of multidrug-resistant Salmonella enterica serovar Albany isolates carried an erm42-carrying integrative and conjugative element, ICE_erm42. Strains carrying ICE_erm42 exhibited high-level resistance to azithromycin and could transfer into phylogenetically distant specie Vibrio cholerae via conjugation.

Multiple-Replicon Resistance Plasmids of Klebsiella Mediate Extensive Dissemination of Antimicrobial Genes

Xue Wang et al.

Summary: The study found that Klebsiella isolates from different wild animals and environments exhibited extensive drug resistance, particularly to ampicillin, cephalosporins, chloramphenicol, levofloxacin, and sulfamethoxazole. Genomic analysis revealed that the resistance plasmids in these isolates carried various antibiotic resistance genes, with multiple-replicon plasmids mainly carrying beta-lactamase genes. Plasmid structure analysis indicated that multiple-replicon plasmids are formed by the fusion of two or more single plasmids.

Analysis of an IncR Plasmid Carrying blaNDM-1 Linked to an Azithromycin Resistance Region in Enterobacter hormaechei Involved in an Outbreak in Quebec

Florence Doualla-Bell et al.

Summary: In the context of rising prevalence of NDM-producing Enterobacterales in Québec, Canada, the genetic environment of blaNDM-1 was investigated in three clinical isolates. Two closely related isolates possessed both a large IncFII conjugative multidrug resistance plasmid and a smaller IncR plasmid containing important antimicrobial resistance genes. This indicates a potential role of these isolates as a reservoir for antibiotic resistance genes.

The IS6 family, a clinically important group of insertion sequences including IS26

Alessandro Varani et al.

Summary: The IS6 family of insertion sequences plays a crucial role in the rearrangement and spread of antibiotic resistance, with particular attention given to IS26 and IS257 for their clinical impact. While limited biochemical data is available on the transposition mechanism of these elements, genetic studies suggest that they may use unexpected mechanisms. This review provides an overview of the family, distribution and impact on host genomes, as well as an analysis of potential transposition pathways.
