Clerodane furanoditerpenoids from the stems of Tinospora sinensis

Summary: One new clerodane-type furanoditerpenoid and nine new nor-clerodane analogs have been isolated from the stems of Tinospora sinensis. The structures and absolute configurations of these compounds were determined using spectroscopic techniques and chemical correlation. Some of the compounds exhibited antiproliferative and alpha-glucosidase inhibitory activities, with compound 10 being a noncompetitive inhibitor and its binding mode to the enzyme further investigated using molecular docking experiments.

Anticancer and Anti-Neuroinflammatory Constituents Isolated from the Roots of Wasabia japonica

Summary: A new compound and 17 previously reported compounds were isolated and identified from the roots of wasabi. The study showed that one of the compounds exhibited powerful inhibitory effects on the proliferation of lung cancer cells and melanoma cells, and the new compound exhibited moderate inhibition activity on nitric oxide production.

Anti-Epstein-Barr Viral Agents from the Medicinal Herb-Derived Fungus Alternaria alstroemeriae Km2286

Summary: This study investigated the liquid-state fermented products produced by the fungal strain Alternaria alstroemeriae Km2286 isolated from the medicinal herb Atriplex maximowicziana Makino. Chromatographic separation resulted in the isolation of compounds 1-9, including four undescribed perylenequinones. Compound 1-6 exhibited antiviral activities against Epstein-Barr virus and compounds 1-4, 6, and 7 showed inhibitory activity of nitric oxide production in lipopolysaccharide-induced microglial BV-2 cells without significant cytotoxicity. This is the first report of perylenequinone-type compounds with potent anti-EBV and anti-neuroinflammatory activities.

Physiologically based pharmacokinetic (PBPK) modeling of piroxicam with regard to CYP2C9 genetic polymorphism

Physiologically based pharmacokinetic (PBPK) modeling of meloxicam in different CYP2C9 genotypes

Summary: The study aimed to develop and validate a physiologically based pharmacokinetic (PBPK) model of meloxicam related to CYP2C9 genetic polymorphism, successfully simulating drug metabolism in different genotypes. The predicted exposures of meloxicam in CYP2C9*1/*3, CYP2C9*1/*13, and CYP2C9*3/*3 genotypes were increased compared to CYP2C9*1/*1 genotype. Through optimization of meloxicam dosing in different genotypes, the study is expected to contribute to reducing the risk of adverse events associated with meloxicam.

Cryptic Biosynthesis of the Berkeleypenostatins from Coculture of Extremophilic Penicillium sp.

Summary: The coculture fermentation of Penicillium fuscum and P. camembertii/clavigerum produced berkeleypenostatins A and E, which showed significant inhibitory effects on the migration of human pancreatic carcinoma cells. The study also revealed that berkeleypenostatins exhibited a potent growth inhibition activity against a wide range of cancer cell lines in the NCI 60 cell five-dose screen.
