ROS/PI3K/Akt and Wnt/β-catenin signalings activate HIF-1α-induced metabolic reprogramming to impart 5-fluorouracil resistance in colorectal cancer

Summary: This study describes the abnormality of glucose metabolism in 5-FU-resistant CRC, and shows that enhanced glycolysis and pentose phosphate pathway are associated with increased HIF-1 alpha expression. HIF-1 alpha-induced glucose metabolic reprogramming imparts 5-FU resistance in CRC. HIF-1 alpha is a potential biomarker for 5-FU-resistant CRC, and targeting HIF-1 alpha in combination with 5-FU may represent an effective therapeutic strategy in 5-FU-resistant CRC.

Atherosclerotic cardiovascular disease and chronic kidney disease are the main causes and they sustain each other. SGLT-2 inhibitors have shown remarkable results in preventing heart failure and renal disease progression, but their benefits for atherosclerotic cardiovascular events are still unclear.

Summary: Excessive production of mitochondrial reactive oxygen species (ROS) in diabetes complications is mainly caused by impaired responses to hypoxia due to the inhibition of hypoxia-inducible factor-1 (HIF-1) by hyperglycemia. Restoring HIF-1 function can attenuate ROS overproduction, protect cells against apoptosis and renal injury in diabetes. These findings suggest that targeting HIF-1 repression could be a potential therapeutic strategy for diabetic complications.

Summary: Diabetes enhances kidney fibrosis by enhancing HIF-1 activation, leading to impaired kidney repair. In diabetic mice, unilateral ureteral obstruction induces more renal fibrosis, apoptosis, and interstitial macrophage infiltration. High glucose-conditioned renal tubular cells show higher expression of fibrosis marker protein under hypoxia, which is mediated by increased HIF-1α expression. Knockout of proximal tubule-specific HIF-1α attenuates fibrosis induced by UUO in diabetic mice kidneys.

MicroRNA-21: A Critical Pathogenic Factor of Diabetic Nephropathy

Summary: This collaborative review provides comprehensive evidence on the role of miR-21 in the development and progression of diabetic nephropathy (DN). Multiple studies have shown that miR-21 is up-regulated in DN and contributes to epithelial-to-mesenchymal transition, extracellular matrix deposition, cytoskeletal remodeling, inflammation, and fibrosis through a complex network with targeted genes and signaling cascades. Further research is needed to confirm the clinicopathological features and molecular mechanisms of miR-21-mediated DN.
