A high-throughput cell-based assay pipeline for the preclinical development of bacterial DsbA inhibitors as antivirulence therapeutics

Anthony D. Verderosa et al.

Summary: The failure of antibiotics has led world health officials to urgently call for the development of new drugs, with a focus on antibacterials against multidrug-resistant Gram-negative pathogens. The development of promising antivirulence drugs is hindered by a lack of standardized preclinical testing methods, slowing down progress in this area. Establishing bacterial cell-based assays for the early evaluation of DsbA inhibitors offers a robust platform for the identification and evaluation of potential treatments.

Salmonella enterica BcfH Is a Trimeric Thioredoxin-Like Bifunctional Enzyme with Both Thiol Oxidase and Disulfide Isomerase Activities

Pramod Subedi et al.

Summary: This study identified and characterized a novel Dsb-like protein, BcfH, from Salmonella that has a trimeric structure and unconventional catalytic sites, contributing to fimbrial biogenesis. The structural and functional flexibility in the TRX-fold protein family was demonstrated, with BcfH showing multiple redox functions mimicking eukaryotic PDI activity in bacteria.

Structural bioinformatic analysis of DsbA proteins and their pathogenicity associated substrates

Summary: The DsbA enzyme, as part of the disulfide bond forming system, plays a crucial role in bacterial virulence factor assembly and is a potential target for new virulence blockers. Despite extensive studies on DSB systems in various bacterial species, little is known about how DsbA oxidoreductases recognize and interact with a wide range of substrates.
