Robust genome editing in adult vascular endothelium by nanoparticle delivery of CRISPR-Cas9 plasmid DNA

Xianming Zhang et al.

Summary: A newly developed nanoparticle-based delivery system has shown high efficiency in genome editing of vascular endothelial cells. This system can selectively decrease protein expression by 80%, achieving a phenotype similar to genetic knockout mice. Furthermore, it has the capability to edit multiple genes or perform both genome editing and transgene expression simultaneously, making it a powerful tool for gene therapy and cardiovascular research.

Endothelial cell heterogeneity and microglia regulons revealed by a pig cell landscape at single-cell level

Fei Wang et al.

Summary: Using single-cell RNA sequencing, researchers generated a comprehensive map of single-cell transcriptomes from various organs of pigs. They identified a high degree of heterogeneity within cells and uncovered several functionally distinct cell types, including a subtype of endothelial cells in adipose tissues. Additionally, they predicted tissue- and cell type-specific communication between endothelial cells and other cell types through specific pathways. Furthermore, analysis of microglia transcriptomes across species revealed an evolutionally conserved regulon.

Summary: This study investigates the therapeutic effect of retinal gene therapy using CRISPR/Cas9-mediated genome editing and knockout applications. By utilizing RNPs composed of msgRNAs and SpCas9, efficacy can be enhanced to achieve targeted gene knockout. Moreover, lipoplex delivery of a mixture of RNP and mRNA has been shown to achieve efficient knockout.

Strategies in the delivery of Cas9 ribonucleoprotein for CRISPR/Cas9 genome editing

Song Zhang et al.

Summary: CRISPR/Cas9 genome editing as a direct delivery method of RNP complex has advantages of transient genome editing and reduced off-target effects, but faces challenges in efficient material delivery. Current Cas9 RNP delivery systems, including physical approaches and synthetic carriers, are detailed in this review, along with their mechanisms and beneficial roles in intracellular delivery, highlighting examples of stimulus-responsive and targeted carriers in RNP delivery. Challenges of current Cas9 RNP delivery systems and perspectives in rational design of next generation materials for this promising field are discussed.
