Fatty Acids: An Insight into the Pathogenesis of Neurodegenerative Diseases and Therapeutic Potential

Diego Julian Vesga-Jimenez et al.

Summary: Palmitic acid (PA), one of the most common lipids in the human body, plays essential functions in brain cells, but its excessive accumulation leads to lipotoxicity and increased susceptibility to neurodegenerative diseases. The activation of toll-like receptors and nuclear factor kappa-light-chain-enhancer of activated B cells pathways, along with the release of pro-inflammatory cytokines, oxidative stress, endoplasmic reticulum stress, and autophagy impairment, are involved in the detrimental effects of PA on brain cells. Oleic acid and linoleic acid have been proposed as potential therapeutic approaches against neurodegenerative diseases by counteracting the negative effects of PA on cells.

Recent studies have shown that specific ceramides can affect multiple metabolic pathways, and reducing selected ceramide pools in obese rodents improves metabolic health.

Differential effects of saturated and unsaturated free fatty acids on ferroptosis in rat ,?-cells

Bastian Kruemmel et al.

Summary: Elevated concentrations of saturated free fatty acids (SFAs) are linked to dysfunction and apoptosis of pancreatic beta-cells. This study found that only ω-6 polyunsaturated fatty acids (PUFAs) induce ferroptosis, while the ω-3 PUFA α-linolenate sensitizes beta-cells to SFA-mediated lipid peroxidation. The prevention of excessive dietary intake of ω-6 PUFAs may be crucial in protecting insulin-producing cells from reactive oxygen species-mediated ferroptosis.

Polydatin prevents lipotoxicity-induced dysfunction in pancreatic β-cells by inhibiting endoplasmic reticulum stress and excessive autophagy

Summary: This study demonstrates that polydatin (PD) protects beta-cells from lipotoxicity-induced dysfunction and apoptosis by inhibiting endoplasmic reticulum stress (ERS) and preventing excessive autophagy. PD treatment enhances insulin secretion and expression of diabetes-associated genes, promoting beta-cell function. In diabetic mice, PD reduces body weight gain, improves dyslipidemia, enhances beta-cell function, and reduces insulin resistance.

Summary: The lipid invasion model proposes that Alzheimer's disease is caused by external influx of free fatty acids and lipid-rich lipoproteins into the brain, following disruption of the blood-brain barrier. This model explains the bioenergetic changes, neuroinflammation, and formation of amyloid plaques and neurofibrillary tangles observed in AD. It also highlights the role of the blood-brain barrier in protecting the brain from external lipid access.

The central role of glutathione peroxidase 4 in the regulation of ferroptosis and its implications for pro-inflammatory cytokine-mediated beta-cell death

The antioxidative enzyme GPx4 is predominantly expressed in beta-cells, and its overexpression as well as the ferroptosis inhibitor ferrostatin-1 can effectively attenuate beta-cell death. Nitric oxide produced during pro-inflammatory cytokine action prevents the induction of ferroptosis, favoring apoptosis as the primary cell death mechanism in beta-cells.
