Electrospun nanofiber scaffolds for the propagation and analysis of breast cancer stem cells in vitro

E. Prieto et al.

Summary: Despite being a major cause of cancer mortality in women, breast cancer continues to have a high rate of relapse after treatment. This study developed a 3D culture platform that mimicked the metastatic tumor ECM to effectively increase CSC population and analyze CSCs. Breast cancer cells grown on the nanofiber scaffolds exhibited characteristics similar to invasive cancer cells, with an increase in breast cancer stem cell population and markers indicating epithelial-mesenchymal transition. These results suggest that the fabricated nanofiber scaffolds effectively mimic the tumor microenvironment and offer a platform to enrich and study CSCs in vitro.

Breast cancer patient-derived scaffolds as a tool to monitor chemotherapy responses in human tumor microenvironments

Maria Carmen Leiva et al.

Summary: The study demonstrated that cell-free patient-derived scaffolds (PDSs) generated from breast cancer samples, recellularized with cancer cell lines, could be used as a model for testing drug responses in breast cancer. The research showed that MCF7 cancer cells cultured in PDSs exhibited increased resistance against frontline chemotherapy drugs compared to traditional two-dimensional cell cultures, and the gene expression of environmentally adapted cancer cells differed depending on the drug used. This approach offers a novel way to study extracellular matrix influences on cancer drug responses and test novel compounds in in vivo-like microenvironments.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

3D In Vitro Models for Investigating the Role of Stiffness in Cancer Invasion

Auxtine Micalet et al.

Summary: This review discusses the role of the physical microenvironment in cancer invasion, finding that increased matrix stiffness may promote cancer invasion, but there are also cases where the opposite occurs. Experimental approaches and interpretations vary, affecting the invasion of cancer differently. The findings can guide future research methods and provide a standardized experimental protocol.
