Human mesenchymal stem cells

Xiaoyong Chen et al.

Summary: Mesenchymal stem cells (MSCs) have potential for cell therapy and tissue regeneration due to their properties, but their heterogeneity poses challenges. The "Human Mesenchymal Stem Cells" guidelines provide standards for quality control and technical specifications, aiming to accelerate the international standardization of MSCs for clinical development and therapeutic applications.

Therapeutic Mesenchymal Stem/Stromal Cells: Value, Challenges and Optimization

Mehdi Najar et al.

Summary: Cellular therapy aims to replace damaged cells and restore the cellular and molecular environments for tissue repair. Mesenchymal stem/stromal cells (MSCs) have therapeutic potential in various diseases. However, the origin and function of MSCs are still debated. The therapeutic properties of MSCs are influenced by their environment, including culture conditions and immunobiology.

Diamond-like carbon coating and surface grafting of osteoprotegerin and alendronate on polyetheretherketone to ameliorate the mechanical performance and osseointegration simultaneously

Junfeng Chen et al.

Summary: Polyetheretherketone (PEEK) is a material used for fabricating bone implants, but its clinical applications are limited by the release of wear debris and poor osseointegration. In this study, a one-stop solution for surface multi-functionalization of PEEK implants is developed by applying a diamond-like carbon (DLC) coating and grafting osteoprotegerin (OPG) and alendronate (ALN) onto the outer surface. This approach improves the wear resistance and biological performance of PEEK implants.

ZnL2-BPs Integrated Bone Scaffold under Sequential Photothermal Mediation: A Win-Win Strategy Delivering Antibacterial Therapy and Fostering Osteogenesis Thereafter

Yuzheng Wu et al.

Summary: This study presents a multifunctional bone implant designed to provide antibacterial therapy and promote bone regeneration through sequential photothermal mediation. The design improves the biosafety of antibacterial photothermal treatment and facilitates osteogenesis through gradual ion release. This strategy may have broader applications in biomedical fields and offers insights into the design of multifunctional biomaterials.

Dexamethasone priming enhances stemness and immunomodulatory property of tissue-specific human mesenchymal stem cells

Sonali Rawat et al.

Summary: The study found that Dex preconditioning can enhance the immunomodulatory properties of hMSCs, reducing the challenge associated with minimal potency and enhancing therapeutic efficacy. Additionally, different tissue-specific hMSCs exhibited varying levels of immunosuppression after Dex preconditioning during mixed lymphocyte reaction, with DP-MSCs and UC-MSCs showing stronger immunosuppressive effects.

Discrepancies on the Role of Oxygen Gradient and Culture Condition on Mesenchymal Stem Cell Fate

Jay R. K. Samal et al.

Summary: Studies have shown that culturing MSCs in a low oxygen environment can upregulate stem cell markers and promote cell proliferation in an undifferentiated state. However, there is ambiguity in the results due to differences in oxygen gradients and culture conditions among different studies. This progress report focuses on conflicting findings related to the application of hypoxic conditions for improving MSC proliferation or differentiation, aiming to decipher the factors influencing MSC characteristics under hypoxia.

Programmed surface on poly(aryl-ether-ether-ketone) initiating immune mediation and fulfilling bone regeneration sequentially

Lingxia Xie et al.

Summary: This study demonstrates the development of a coated polymer material, PEEK, for immune modulation in different stages of bone healing by rapid release of IL-10 and slow delivery of DEX. The sequential immune-mediated process effectively reduces inflammation, promotes M2 polarization of macrophages, and upregulates autophagy-related factors, ultimately facilitating bone regeneration. This innovative approach paves the way for the fabrication of advanced biomaterials for tissue engineering and immunological therapeutics.
