Challenges in Treating Genodermatoses: New Therapies at the Horizon

Summary: Genodermatoses are rare inherited skin diseases that often affect other organs, causing significant impacts on patients' health and well-being. Advances in molecular genetics and translational research have provided opportunities for innovative treatments, including intervention in cell-signaling pathways, suppression of inflammation, and correction of genetic defects. These technologies vary in terms of personalized, short-term, and permanent therapeutic effects. Prevention of transmission to future generations is also an important goal for severe genodermatoses.

Summary: This article describes a detailed protocol for generating hair-bearing skin tissue from human pluripotent stem cells. By using defined culture conditions in a three-dimensional in vitro culture system, skin organoids with hair follicles can be obtained after 60 days and reach full complexity after approximately 130 days.

Characterization of the epidermal-dermal junction in hiPSC-derived skin organoids

Application of an iPSC-Derived Organoid Model for Localized Scleroderma Therapy

It was found that the application of human-induced pluripotent stem cell-derived epithelial and mesenchymal (EM) organoids in a 3D culture system can reduce skin fibrosis and promote sweat gland and blood vessel regeneration.

Generation of Keratinocytes from Human Induced Pluripotent Stem Cells Under Defined Culture Conditions

Summary: A simplified method was proposed in the study to efficiently generate large numbers of uniformly differentiated keratinocytes from hiPSCs in less than 4 weeks, with high homogeneity and low cell death rate. The hiPSC-derived keratinocytes, which can be maintained in a proliferative state and terminally differentiated under high calcium conditions, consistently formed organotypic 3D skin equivalents when cocultured with hiPSC-derived fibroblasts. Keratinocytes generated by this method are a viable source of cells for downstream applications.
