ALD-R491 regulates vimentin filament stability and solubility, cell contractile force, cell migration speed and directionality

Metastasising Fibroblasts Show an HDAC6-Dependent Increase in Migration Speed and Loss of Directionality Linked to Major Changes in the Vimentin Interactome

Summary: This study investigated the regulatory role of vimentin in the motility of metastasizing fibroblasts. Through various techniques including STED super-resolution microscopy, live-cell imaging and quantitative proteomics, the researchers discovered that oncogene-expressing and metastasizing fibroblasts exhibited altered cell shape, cell spreading, migration speed, and migration directionality compared to normal control cells. They also identified 555 proteins in the vimentin interactome and observed changes in protein levels that accompany the defective motility of metastasizing cells. Inhibition of HDAC6 was found to reverse these motility defects and alter vimentin organization. These findings suggest that oncogenes may act through HDAC6 to regulate the interaction between vimentin and cytoskeletal components, leading to defective motility in metastasizing cells.

A Small Vimentin-Binding Molecule Blocks Cancer Exosome Release and Reduces Cancer Cell Mobility

Summary: Vimentin-binding compound R491 induces intracellular vacuolization in cancer cells and significantly reduces exosome release. Cells with lower vimentin levels have fewer exosomes, while R491 can decrease cancer cell migration and invasion in vitro and reduce exosome levels in mice. These findings suggest potential applications for inhibiting cancer invasive growth by targeting vimentin.
