Enhancement strategies for transdermal drug delivery systems: current trends and applications

Elucidating the Impact of Surfactants on the Performance of Dissolving Microneedle Array Patches

Soluplus®-based dissolving microarray patches loaded with colchicine: towards a minimally invasive treatment and management of gout

Summary: Considered as one of the most common inflammatory arthritis, gout is characterised by a sudden onset of severe joint pain. As the first-line drug of choice used in treating acute gout, colchicine (CLC) is hindered by poor gastrointestinal permeability as well as unfavourable gastrointestinal side effects. Herein, we present, for the first time, the preparation of microarray array patches (MAPs) made of a polymeric solubiliser, Soluplus (R), loaded with CLC for its systemic delivery. The fabricated MAPs displayed acceptable mechanical properties and were capable of being inserted into the skin to a depth of approximate to 500 mu m in full thickness ex vivo neonatal porcine skin, as evidenced by optical coherence tomography. In vitro dermatokinetic studies utilising full thickness neonatal porcine skin demonstrated that the CLC-loaded MAPs delivered CLC across all skin strata, resulting in a delivery efficiency of 73% after 24 hours. Furthermore, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) and cell proliferation assays along with LIVE/DEAD (TM) staining on the 3T3-L1 cell line showed that the MAP formulation displayed minimal toxicity, with acceptable biocompatibility. Lastly, the anti-inflammatory properties of the formulation were evaluated using a THP-1 macrophage cell line. It was shown that treatment of THP-1 macrophages that are exposed to lipopolysaccharide (LPS) with CLC-loaded MAPs caused a significant (p < 0.05) reduction of TNF-alpha production, a pro-inflammatory cytokine typically associated with the early onset of acute gout. Accordingly, CLC-loaded MAPs could represent a new minimally-invasive alternative strategy for management of acute gout.

Dissolving microneedle patches loaded with amphotericin B microparticles for localised and sustained intradermal delivery: Potential for enhanced treatment of cutaneous fungal infections Ke

Summary: Fungal infections are a global issue affecting millions of individuals, with conventional treatments often proving ineffective due to low drug bioavailability and drug resistance. Amphotericin B (AmB) loaded dissolving microneedle patches were introduced for localized and long-acting intradermal delivery to treat cutaneous fungal infections, demonstrating significant inhibition and killing of Candida albicans. Moreover, pharmacokinetic studies showed lower systemic exposure and reduced systemic side effects compared to intravenous injection, highlighting the potential of this novel drug delivery system.

Retaining Skin Barrier Function Properties of the Stratum Corneum with Components of the Natural Moisturizing Factor-A Randomized, Placebo-Controlled Double-Blind In Vivo Study

Summary: This study investigated the effects of a topically applied formulation containing components of natural moisturizing factor (NMF) on barrier-related parameters of the stratum corneum (SC). It was found that the formulation disrupted the equilibrium of water, NMF, and lipids in the SC, but adverse effects could be mitigated by a simplified NMF supplementation. Furthermore, the supplement unexpectedly prevented intercellular lipids (ICL) disorganization at selected depths, and limited lateral disorganization of ICL to the upper 20% SC depth in the long term. Further research is needed to understand the interaction of these factors in the SC for the development of topical treatments for skin barrier-related diseases.
